The Expression And Significance Of CD40LmRNA, PI3KmRNA And AKTmRNA In Bone Marrow Of Patients With Acute Leukemia (M2,M5) | | Posted on:2017-04-02 | Degree:Master | Type:Thesis | | Country:China | Candidate:M Liu | Full Text:PDF | | GTID:2284330503980315 | Subject:Internal Medicine | | Abstract/Summary: | | | Objective:By detecting the expression of CD40 Lm RNA, PI3 Km RNA and AKTm RNA in acute myeloid leukemia(M2, M5) before and after chemotherapyto explore the value of CD40 Land PI3K/AKT signaling pathway in the pathogenesis and treatment of acute myeloid leukemia.Method:All patients were divided into three groups:before chemotherapy group, after chemotherapy group and control group. Real-time fluorescence quantitative PCR was used to detect the expression of CD40 Lm RNA,PI3 Km RNA and AKTm RNA in bone marrow tissue of 31 patients with newly diagnosed AML(M2, M5) before and after chemotherapy; 20 cases of non-malignant hematologic diseases(iron deficiency anemia, megaloblastic anemia) were collected as control group.The expressions of CD40 Lm RNA,PI3 Km RNA and AKTm RNA in bone marrow tissue were compared between all three groups.Results: 1)Before chemotherapy, the expression of CD40 Lm RNA was lower than control group and after chemotherapy group(P<0.05).The expressions of PI3 Km RNA and AKTm RNA were higher than control group and after chemotherapy group(P<0.05). 2)There were 18 cases achieved remission(including 17 cases of complete remission and1 case of partial remission). We compared the expression of CD40 Lm RNAof all patients who had achieved remission included complete and partial remission of before and after chemotherapy, it was increased in after chemotherapy group(P<0.05). And the expressions of PI3 Km RNA and AKTm RNA in after chemotherapy group were decreased(P<0.05).3) After chemotherapy, there were 13 patients who did not relieve, the expressions of CD40 Lm RNA,PI3 Km RNA and AKTm RNA of all patients who had not achieved remission were not significantly different in both before and after chemotherapy groups(P>0.05).Conclusion:1) In AML(M2, M5) patients, before chemotherapy, the expression of CD40 Lm RNA was decreased and the expressions of AKTm RNA and PI3 Km RNA were increased. In AML(M2, M5) patients who achieved remission,the expression of CD40 Lm RNA after chemotherapy group was higher and the expressions of PI3 Km RNA and AKTm RNA were lower than before chemotherapy group.2) In AML(M2, M5) patients, low expression of CD40 L and activation of PI3K/AKT pathway mightrelate to pathogenesis and therapeutic efficacy of AML. | | Keywords/Search Tags: | acute myeloid leukemia, CD40L, PI3K, AKT | | Related items |
