Objective1. Preliminary study of Huoxue Dingxuan Capsule on serum intervention CSA rat model and the effect of TNF-α, IL-1βand IL-6;2. Clear Huoxue Dingxuan Capsule on CSA model rat peripheral blood T lymphocyte subsets CD3~+, CD4~+, CD8~+ count ratio;3. For Huoxue Dingxuan capsules promotion and provide experimental basis for clinical treatment of CSA.MethodWill be 90 SPF grade male Wistar rats, 12 months old, weighing 200-250 g, adaptive feeding after a week, were randomly assigned for two groups: the a group as the blank group(Group A, n=15); another group of modules to build. After modeling success then were randomly divided into five groups: model group(Group B, n=15), promoting blood circulation capsule glare low dose group(Group C, n=15) and promoting blood circulation capsule glare dose group(Group D, n=15), promoting blood circulation capsule glare in high dose group(Group E, n=15), control group(Group F, n=15). Four weeks after rat model was made successfully, packet delivery, blank group and model group with equal dose physiological saline gavage, control group for Jingfukang medicine to equivalent dose conversion dilution after intragastric administration, continuous administration for 6 weeks. ELISA method to detect CSA model rats serum TNF-α, leukocyte mediated IL-6 and leukocyte mediated IL-1β expression, flow cytometry detection and peripheral blood T lymphocyte subsets CD3~+, CD4~+, CD8~+ cell counts the proportion.Result1. After modeling, rats of model group CSA generally compared with the control group, a decrease in the amount of diet, stool less viscous, messy hair color, reduce the volume of activities, blood flow compared with the blank group decreased significantly(P < 0.05).2. Comparison between before and after the intervention in treatment group compared to Huoxue Dingxuan capsule drug intervention before making model rats and the normal rats, the serum level of TNF-α, IL-6 and IL-1β content increased significantly(P<0.05); intervention, Huoxue Dingxuan capsule treatment group rats serum TNF-α, IL-6 and IL-1β were significantly lower than that in the model group(P<0.05), but with Jingfukang group, the difference was not statistically significant(P > 0.05). And the group before intervention were compared, Huoxue Dingxuan capsule treatment group intervention serum TNF-α, IL-6 and IL-1β content decreased(P<0.01). Huoxue Dingxuan capsule(low, medium and high dose group, the difference was not statistically significant(P> 0.05).3. Compared the contrast, CSA model rats of CD3~+, CD4~+ count was significantly increased and CD8~+counts decreased CD4~+/CD8~+ ratio increased, the difference is significant(P< 0.01). Drug intervention, Huoxue Dingxuan capsule treatment group and intervention of contrast, CD3~+, CD4~+ cell count decreased CD4~+/CD8~+ ratio decreased, the difference was statistically significant(P<0.05), and the intervention group and blank group were compared, the difference was statistically significant(P<0.05); Huoxue Dingxuan capsule low and high dose group after the intervention, the difference was not statistically significant(P>0.05); Huoxue Dingxuan capsule group and Jingfukang group, the difference was not statistically significant(P > 0.05).Conclusion1. The CSA model rats, serum immune cell factor TNF-α, IL-6, IL-1βand CD3, CD4~+ content increased, there is a certain relationship between the change of CSA may prove pathogenesis and immune cell factors.CSA model rats treated with Huoxue Capsule glare drug intervention, the serum TNF-α, IL-6 and IL-1βcontent decreased significantly, proving by promoting blood circulation capsule glare can effective intervention of CSA disease caused by the changes of immune cytokine. The molecular mechanism may be related to the therapeutic effect of Huoxue Dingxuan capsules.2. CSA model rats treated with Huoxue Capsule glare drug intervention, peripheral blood CD3~+, CD4~+ expression quantity and ratio of CD4~+ / CD8~+ obviously than pre decreased, prompt Huoxue Dingxuan capsule by improving the immunity of rat model of CSA to achieve the purpose of CSA treatment.
