| Echinacea purpurea(Echinacea purpurea L.) is also known as Echinacea, perennial herbaceous plants of Echinacea Moench, toghter with Echinacea angustifolia(E.angustioflia) and Echinacea(E.pallia) being the higher value type used in traditional Chinese medicine. Echinacea was a folk herbal medicine in America and Europe for hundreds of years of history. In recent years, Echinacea attracted extensive attention from international scholars for its significant functions in anti inflammation, immune regulation, and supplimentary immune. In this paper, Echinacea purpurea leaves were used as the tested materials to carry out echinacoside component extraction, purification and determination process and anti-aging research. The results were as follows:1. Extraction process: Echinacoside component was determined by reversed-phase liquid chromatography.The extent of grinding Echinacea, solvent species, ethanol concentration, solid-liquid ratio and extraction times of factors were used as L9(34) orthogonal experiment list to explore the optimum extraction of Echinacoside. The optimal purification conditions of was selected by contrast of Echinacoside content purified by three macroporous resins of HPD-100, D-101 and AB-8 to extract a complete and optimal Echinacoside purification process.The results showed that echinacea was ground to 60 mesh, using 70% ethanol as an extraction liquid, solid-liquid ratio 1:10, extracting twice.2. The qualitative and quantitative methods and purification methods: The extracting solution was absorbed with HPD-100 macroreticular resin process for 24 hours, used ultrapure water to clean impurities and 50% ethanol as the eluent, to dry with electric heating blower box, and measured by HPLC Echinacosideto determine the qualitative and quantitative. HPLC conditions were Discovery C18 column(25cm*4.6mm, 5μm), 34℃, 10μl injection volume, and mobile phase: 0.1% formic acid: methanol--73.5: 26.5, continued for 17 min, then rapid(<3min) change the mobile phase to 0.1% formic acid: methanol--70.5: 29.5, continued for 15 min, set a flow rate of 0.9mL / min, checking at 330 nm wavelength as the most suitable conditions.3. Anti aging test: the paper established conventional aging mice mode, D- amino galactose method to induce aging animals. And the mice were divided into the control group, the model group and Echinacoside group. The mice of Echinacoside group were intragastric with echinacoside to detect related aging indicators, to investigate the mechanism of anti- aging of echinacoside by collecting data and analyzing. The results showed that 1) Echinacoside increased the compensatory of the immune organ in mice and INF-γ, and decreased IL-6 levels in serum. The occurrence of aging was delayed by regulating the pathways of cellular immunity. 2) At the same time, by inhibiting the expression of TGF-β1 and p53 gene in kidney, Echinacoside delayed cell aging and reduced the symptoms of aging mice.4. Antibacterial experiment: double dilution method was used to test the minimum inhibitory concentration of Echinacea essential oil for several bacteria or fungi. The minimum inhibitory concentration for Escherichia coli., Staphylococcus aureus, and Saccharmoyces cerevisiae were13%, 6%, and3% respectively. And it is not obvious to Monascus purpureus Went.By diffusion method, potassium sorbate was used as contrast to compare with antibacterial activity of echinacea essential oil and chamomile essential oil. The results showed that potassium sorbate was the optimal, and echinacea essential oil was the next when concentration was 10%. |
