Objective: To establish HPLC fingerprints to ensure the quality of the source of Astragalus and Pueraria herbs, monitoring Tangshen Pellets preparation process to control its quality.Method:(1) Using the testing method of HPLC fingerprint of astragalus herbs,16 different habitats Astragalus were measured, calibration common peaks, to determine the reference peak, to calculate relative peak areas and relative retention times of the common peaks, to Calculate the area of the common peaks,the fingerprint similarity analysis and cluster analysis.(2)using the testing method of HPLC fingerprint of Pueraria herbs, 16 different habitats Pueraria herbs were measured, calibration common peaks, to determine the reference peak, to calculate relative peak areas and relative retention times of the common peaks,to Calculate the area of the common peaks,the fingerprint similarity a nalysis and cluster analysis.(3) HPLC fingerprint method for the determination of Tangshen Pellets(composed mainly containing Pueraria and astragalus), and for ten batches Tangshen Extract and three batches of Tangshen pellets were measured, respectively, to determine both the common peaks, calculate the relative retention time and relative peak area to calculate the similarity of fingerprints; Tangshen Pellets common peaks were assigned analysis.Results:(1) 14 Astragalus herbs common peaks were calibrated, peak area greater number 1 peak as reference peak; Relative retention less time difference, the larger the relative difference in peak area; the results of fingerprint similarity analysis and cluster analysis is consistent; small regional relations between the similarity and cluster analysis of samples, these herbs’ total area of common peaks and better quality herbs have a certain relationship.(2)13 Pueraria herbs total peaks were calibrated, peak area greater number 3 peak as reference peak; relative retention less time difference, the larger the relative difference in peak area; the results of fingerprint similarity analysis and cluster analysis is inconsistent; small regional relations between the similarity and cluster analysis of samples, small regional relations cluster analysis, similarity with the sample.(3)The number of total peaks of ten batches of Tangshen Extract Fingerprints is 22; The number of total peaks of three batches of Tangshen Pellets Fingerprints is 22; there are 20 same total peaks peaks in the Tangshen Extract and Tangshen Pellets Fingerprints, the individual components are different; respectively, the 5th peak is reference peak; relative retention time is few difference between samples, relative peak area is in larger differences; the fingerprints similarity of ten batches of Tangshen Extract and batches of Tangshen Pellets is high, indicating that the preparation process is stable and reliable. Tangshen Pellets 1 to 12,15,16 and 18 common peaks derived from Pueraria, 13 common peaks derived from astragalus; 17 common peaks derived from astragalus and Pueraria.Conclusion:(1)the HPLC fingerprint of Astragalus measurement method established by the Institute is simple, accurate, reliable, and can be used as a reference medicinal Astragalus quality control.(2)the HPLC fingerprint of Pueraria measurement method established by the Institute is scientific,accurate, reliable, and can be used as a reference medicinal Pueraria quality control.(3) the HPLC fingerprint of Tangshen pellets measurement method established by the Institute is scientific, accurate, reliable, and can be used as a reference medicinal Tangshen pellets quality control.
