| CURCUMAE RHIZOMA,the dried rhizomes of Zingiberaceae Curcuma phaeocaulis Val., Curcuma kwangsiensis S.G.Lee et C.F.Liang and Curcuma wenyujin Y.H.Chen et C.Ling. Curcuma kwangsiensis is the famous-regio drug in Guangxi and distributed in middle and southern area of Guangxi.Our research group introduced and cultivated the Curcuma kwangsiensis from genuine producing area of Guangxi to Guangdong region,which to provide the basis for the introduction and cultivation of Curcuma kwangsiensis and expand the planting area. In this paper, CURCUMAE RHIZOMA which from different regions were focused on researching their quality. The essential oil was determined by steam distillation and analyzed by using GC-MS. In this work, methods for the determination of germacrone,β-elemene and total polysaccharides in CURCUMAE RHIZOMA were established.Aims:To establish the menthods of quality evaluation of CURCUMAE RHIZOMA and compare the different of essential oil contents and other chemical compositions of CURCUMAE RHIZOMA from different producing area and cultivars.Methods:1. The quantitation of essential oil: According to the appendix XD below items of2010 version volume one of “Chinese Pharmacopoeiaâ€, this work determined the essential oil contents of the collected samples of Curcuma kwangsiensis.2. GC-MS analysis: The essential oils were analyzed by GC-MS.To confirm the main component of essential oil by comprehensive analysis, according to the computer retrieval and parsing library system of the NIST2.0 spectrum, and relevant literature and data. The relative content of compositions was determined by the method of area normalization.3. Quantitation: Methods for the determination of germacrone, β-elemene and total polysaccharides in CURCUMAE RHIZOMA were established by high performance liquid chromatography, gas chromatography and ultraviolet spectrophotometry.Results:1. The quantitation of essential oil: The content of essential oil in the range of0.71%~2.0%. Among the 11 batch of samples,only 6 batches up to specification and the qualified rate was 55%.2. GC-MS analysis: More than 40 compounds which included 22 common compounds were identified through GC-MS analysis.Germacrone, β-elemene, curdione with antitumor activity, antiviral and antibacterial were also detected. The cluster analysis and principal component analysis results were good which could get different producing area and cultivars of CURCUMAE RHIZOMA together.3. Quantitation: The calibration curves were linear in the ranges of2~52μg/m L(R2=0.9997) for germacrone, 0.04224~0.8448mg/m L(R2=0.9996) forβ-elemene, 0.02~0.10mg/m L(R2=0.9992) for total polysaccharides respectively. The average recoveries were 99.37% with RSD 2.21% for germacrone, 99.68% with RSD2.37% for β-elemene, 99.10% with RSD 2.30% for total polysaccharides respectively.Conclusions:1. The quantitation of essential oil: The result showed that part of the batches was not in conformity with the provisions, and it was significantly different among different batches of essential oil contents of CURCUMAE RHIZOMA.2. GC-MS analysis: The content and kind of chemical compositions of essential oil in CURCUMAE RHIZOMA which from different producing area and cultivars were different.The essential oil was mainly made of monoterpenoids and sesquiterpenoids.3. Quantitation: Methods for the determination of the three compositions in CURCUMAE RHIZOMA were established by high performance liquid chromatography,gas chromatography and ultraviolet spectrophotometry. These methods can be used for the quality control of CURCUMAE RHIZOMA were simple and accurate. |
