Objective: Stress refers to the non-specific systemic reactions of the body caused by various internal and external environmental factors. In recent years, various kinds of living environment and social pressure stress factors have been important influences on human health, and they gradually become one of the main factors that threaten human health. The cold environment, as one of them, has a wide influence on human physiological functions, including thermal balance, energy metabolism, water and salt metabolism, neuroendocrine system and cardiovascular system, etc. Therefore, screening of the effective drugs to improve or even eliminate the adverse reactions of the body caused by chronic cold stress is of great significance. Studies have shown that Rhodiola can adjust immune functions of normal human under the condition of low temperature, and enhance the abilities of cold and stress resistance. Under the guidance of the theories of TCM, nuclear magnetic resonance(NMR)-based metabonomics was applied to study the treatment effect of Rhodiola on protecting rats induced by chronic cold stress on the whole, and to clarify the metabolic regulation mechanism. This research supplies useful theoretical basis for the prevention and treatment of chronic cold stress.Methods: In present study, we collected the plasma, urine, feces and large intestine tissue samples of control, chronic cold stress and Rhodiola treatment(5 g·kg-1·d-1) groups. And then we did the metabonomic research and analysed the metabolic regulation of Rhodiola, to screen out the potential metabolic markers and reveal the metabolic regulation mechanism of Rhodiola in protecting the rats induced by chronic cold stress. At the same time, the serum biochemical parameters of rats were measured.Results: Biochemical determination of the serum results show that the disorders in organism and the hepatic and renal dysfunction induced by chronic cold stress, and the body could reover from a large extent after Rhodiola treatment, with statistical significance. Rhodiola can regulate the metabolic profiles of plasma, urine, feces and large intestine tissues of chronic cold stress rats, and make the metabolic characteristics tend to the normal group from a hostic view.(1) Compared with the model group, Rhodiola decoction can increase the level of glycine and decrease the level of lactate, acetoacetate, creatine, creatinine, choline metabolites(GPC, PC and betaine), TMAO and taurine in plasma.(2) Rhodiola decoction can also inhibit the increase of TCA cycle intermediates(2-oxoglutarate and citrate), acetoacetate, taurine, TMAO and betaine levels in the urine samples which caused by chronic cold stress and elevate the level of tryptophan and creatinine.(3) Rhodiola decoction can decrease the level of lactate, choline, glutamine, glutamate, trimethylamine, TMAO, taurine, hippurate and cadaverine in the fecal extracts which induced by chronic cold stress, and increase the level of SCFAs(butyrate, propionate acetate and valerate).(4) Rhodiola decoction can also effectively inhibit the increase of lactate, choline, glutamate, glutamine, trimethylamine, TMAO, taurine and hippurate in large intestine tissue extracts, and elevate the acetate return them to normal.Conclusion: In the dissertation, the research involves metabonomics and biochemical determination to ascertain the effect of Rhodiola decoction on protecting rats induced by chronic cold stress. Rhodiola can regulate enery metabolism, amino acid metabolism, the glucolipid metabolism and intestinal flora metabolism to relief the metabolism disorders of the plasma, urine, feces and large intestine tissues in chronic cold stress rats. The study elucidated the metabolic regulation mechanism of the protection of Rhodiola in chronic cold stress rats, suppling useful basis for clinical application, and it provided the theoretical basis for the prevention and treatment of chronic cold stress.
