Objective: To observe the ultrastructure changes of ramatroban on podocyte apoptosis induced by puromycin aminonucleoside(PAN).Methods: The conditionally immortalized murine podocytes were randomly divided into four groups: podocyte control group( CON group); podocyte exposed to PAN(100mg/ml) for 24h(PAN group);the podocytes were pretreated for 1h with ramatroban(100 μg/m L) before PAN(100mg/ml) exposure(RA+PAN group);podocyte exposed to ramatroban(100 μg/m L)(RA group). They were collected respectively at 24 and 48 h.The level of podocytes apoptosis was detected by FITC-Annexin V/PI assay under fluorescence microscope. The ultrastructure changes of podocyte morphology were observed with transmission electron microscope.Results: In PAN group,the apoptosis was significantly increased both at 24 h [(11.75±3.01)%] and 48 h [(32.42±6.46)%] compared with the control group(2.67±1.49)%( P < 0. 01),while the apoptosis of RA+PAN groups was obvious decreased both at 24h(6.42±2.27)% and 48h(18.83±4.13)% compared with PAN group( P < 0. 05).No obvious apoptosis was observed in the RA group.Compared with PAN group, the cell ultrastructural damage was significantly reduced after treatment with ramatroban at 24 and 48 h.TEM displayed that cytoplasmic vacuolar degeneration and apoptotic bodies obviously decreased.Conclusion:Ramatroban can reduce PAN-induced podocyte apoptosis rate and cell injury.
