Objective:By comparing the healthy rats and diabetic rats myocardial mitochondria fusion protein 1(Mfn1) the difference of expression, to explore the change of myocardial mitochondria fusion protein in the state of diabetes,and diabetic rats myocardial with reperfusion processing and sevoflurane post-conditioning respectively, detection of myocardial mitochondria Mfn1 expression, discusses diabetes rat myocardial mitochondria Mfn1 expression in reperfusion processing and sevoflurane post-conditioning of change, reveals the effects of sevoflurane post-conditioning in diabetic rats myocardial reperfusion injury.Methods:Sixty clean healthy adult Sprague-Dawley rats, weight 200~240g, using a random number table method, it is divided into healthy control group(S group), control diabetes group(DS group), ischemia-reperfusion group with the diabetes mellitus(DI/R group)and sevoflurane postconditioning diabetic group(DSev group). The preparation of diabetic rats model using disposable large(65 mg/kg) chain of urea with streptozotocin in abdominal cavity injection method. Myocardial ischemia-reperfusion injury model by left anterior descending coronary artery(LAD) ligation 30 min, loose line 120 min reperfusion method. S and DS group only for threading, DI/R group and DSev group ligation LAD complete ischemia reperfusion, which the sevoflurane(2.4 vol %) was inhaled for 5 minutes starts from 29 min of ischemia in DSev group. At the end ofreperfusion, rats were sacrificed and cardiac tissues were obtained. The myocardial infarction size was measured by TTC staining method. The determination of myocardial cell apoptosis index(AI) using TUNEL method. The expression of myocardial tissue mitochondrial Mfn1 protein levels was determined by immune histochemical method.Results:Comparison the area of myocardial infarction, compared with S group, myocardial infarction size difference is so small which is not significant in statistical study in the DS group(P>0.05). the area of myocardial infarction size was increased in the DI/R group and DSev group(P<0.05). Compared with the DS group, the area of myocardial infarction was increased in DI/R group and DSev group(P<0.05). Compared with the DI/R group, DSev group of myocardial infaraction area has no meaning in the study of statistical difference(P>0.05).The comparison of myocardial apoptosis index :compared with S group, the myocardial apoptosis index was significant increased in the DS group、DI/R group and DSev group(P<0.05). Compared with the DS group, the myocardial apoptosis index was significant increased, in DI/R group and DSev group(P<0.05). Compared with the DI/R group,DSev group myocardial apoptosis index difference in statistical study have no significance(P>0.05).The comparison of mitochondrial fusion protein expression(Mfn1):compared with S group, the expression of myocardial mitochondrial Mfn1 protein decreased significantly in the DS group、DI/R group and DSev group(P<0.05). Compared with the DS group, the expression of myocardial mitochondrial Mfn1 protein was obviously decreased in the DI/R group and DSev group(P<0.05). Compared with the DI/R group the expression difference of myocardial mitochondrial Mfn1 protein in the DSev group is so small which has no meaning in statistical study.(P>0.05).Conclusion:The expression of diabetic rats myocardial Mfn1 than normal rats decreased significantly, diabetic rats myocardial reperfusion injury can futher reduce the expression of mitochondria Mfn1. Sevoflurane postconditioning does not apparent affect the expression of Mfn1 in diabetes rats which was going with reperfusion injury...
