Objectives:1.To investigate the effect of sitagliptin on high glucose-induced rat tubular epithelial cell apoptosis.2.By observing the change of protein JNK on injury cells,to explore the mechanism of sitagliptin Methods:Tubular epithelial cells were cultured and divided randomly into four groups:⑴Control, ⑵High glucose(HG in short),⑶sitagliptin(ST in short),⑷JNK inhibitor(JNK in short).The apoptosis of cells was determined using flow cytometry;The phospho-JNK was determined using Western blot analysis and RT-PCR.Results:1.flow cytometry show: compared with the control group, the apoptosis was increased in HG group; compared with the HG group, the apoptosis of ST and JNK group was decreased, the difference was statistically significant(P<0.05).2.RT-PCR show: compared with the control group, the expression of pJNK was increased in high glucose group; compared with the high glucose group, the pJNK of ST and JNK group was lower,the difference was statistically significant(P<0.05).3.Western Blot show: compared with the control group, the expression of pJNK was increased in HG group; compared with the HG group, the pJNK of ST and JNKgroup was lower,the difference was statistically significant(P<0.05).Conclusions:Sitagliptin can inhibit glucose-induced rat tubular epithelial cells apoptosis.The mechanism of this action probably involves that sitagliptin blocks JNK signaling pathway.
