Objective:(1). To study the expression of WT1 protein in breast cancer;(2). To discuss the differential expression of WT1 protein in different subtypes of breast cancer, and the relationship between the expression of WT1 protein and lymph node metastasis;(3). This study provides a basis for the further study of the role of WT1 in the occurrence and development of breast cancer, and provides a idea for the development of clinical work in breast cancer. Methods:1.Selection the breast cancer tissue paraffin blocks which have diagnosed by clinical and pathological mechanism in breast cancer patients, and the selected patients were not any intervention treatment before surgery.2.The expression of WT1 protein in breast cancer was detected by PV-9000 two step immunohistochemical method, and the experimental results were evaluated with the intensity of staining and the positive proportion of cancer cells.3. Statistical analysis: Using SPSS16.0 statistical software to the experimental data’s statistics and analysis. According to the different types of indicators, continuous data with mean and standard deviation to describe, among the three groups compared with the F test, further comparisons with SNK test; classification data use case number and rate description, among the three groups are compared with the 2? test, further comparisons with 2? segmentation; categorical variables use the Spearman rank correlation analysis. The P<0.05 test is statistically significant. Results:WT1 protein was highly expressed in breast cancer, and the expression rate was 52%. In the three groups of different subtypes, HER-2 subtype expression rate was 81.2%, TNBC subtype expression rate was 41.2%, and Luminal subtype expression rate was 35.3%. The expression of WT1 protein in the three different subtypes of breast cancer was statistically significant.(χ2=7.383,P=0.025). Between pairwise comparisons, HER-2 vs Luminal(χ2 =7.993, P = 0.046), TNBC vs Luminal(χ2=8.562 P = 0.037),and the difference of results was statistically significant. In different axillary lymph node positive group, WT1 protein expression differences between four groups was statistically significant(Fisher=16.199, P= 0.018). Between pairwise comparisons, N1 mi group vs 2-3 group(Fisher=9.002, P=0.003), N1 mi group vs is ≥4 group(Fisher=10.251, P = 0.001), the difference of results was statistically significant. And WT1 protein expression and axillary lymph node metastasis showed significant correlation(r = 0.665, P < 0.0001). Conclusion:(1). WT1 protein may play an important role in the occurrence and development of breast cancer;(2). WT1 protein may improve the invasion and metastasis of breast cancer cells, resulting in the enhancement of its malignant degree.
