Objective:Our previousstudy found that in the position of mouse’sShenque acupoint there was a lump of fatty tissue.Sca-1--positive cells was found during immunohisto chemical staining of these fats. And its distribution was agreed with Ren Mai.This study will compare the Shenque acupoint’fatty tissue with ordinary adipose tissue. Then analysis of both morphology and the amount of Sca-1--positive cells will be done to see wheather Shenque cellulite is specific. Next step, stepcreating a simple and typical animal disease model to simulate Shenque applicator therapy and explore the relationship between Shenqueadipose tissue and its functionin during the treatment throughShenque acupoint on animal level. While from Shenque adipose tissue pluripotent stem cells were isolated, cultured and identified. Our study will provides a new direction and lay an experimental basis for further study. Methods:Stainthe Shenque cellulite tissueand groin’s ordinary adipose tissue with immunohistochemistry technique and compares the morphology and Sca-1--positive cells. Injection of LPS was used to make a mouse fever model. Six groups were designed to explore S henque cellulite’s function. Shenque cellulite waspeeled under a dissecting microscope to get the pluripotent stem cells in Shenque cellulite.The cells were cultured using the method of tissue adherentculture. The surface markers of the cultured cells were analysed by flow cytometry. Adipogenic and osteogenic differentiation medium were prepared to induce stem cells to adipogenic and osteogenic differentiation. After adipogenicdifferentiation the lipid droplets in the cytoplasm was stained with oil red O. After osteogenic differentiation, thecalcium deposition was stained using alizarin red. Results and conclusion:1.Mouse’ Shenque acupoint cellulite has denserstructure and contains more cells than common inguinal adipose tissue. Mouse’ Shenque acupoint cellulite was rich in Sca-1--positive cells.2.LPS can induce mouse stable fever which was self- limiting. After Shenq ue paste treatment the temperature dropped tonormal level but when Shenque c ellulite was removed, the cooling paste had no effect. So we think that Shenqu e cellulite has a very important role in Shenque acupoint treatment process.3.Pluripotent stem cells can be extracted from Shenque adipose tissue. The se stem cells were positive ofSca-1, CD105, CD44, and negative of CD31, C D45, CD166 and CD34. The adipogenic and osteogenic differentiation test prove dthat Shenque pluripotent stem cells have unique plasticity and can differentiate in a variety of directions.
