Objective : To discuss mechanisms and associated protective factors of upper gastrointestinal mucosal lesions(AGMLs) following traumatic brain injury(TBI) on mice, to explore the value of heat shock protein 70 on mice TBI related AGMLs, and to evaluate prophylactic efficacy of Geranylgeranylacetone(GGA) administrated in TBI of mice.To probe reasonable measures to effectively prevent and therapy TBI related AGMLs.Methods:TBI related AGMLs was induced via the improved Feeney impact model. Forty adult male Balb/c mice were randomly divied into sham(A),TBI(B),TBI+GGA(C)administered,TBI+saline(D) groups. GGA(800 mg/kg) was administered via oral tube beginning before the model was built 3 hours.The expression of HSP70 protein of brain and gastric mucosa were determined by immunohistochemistry, and the apoptotic index were detected by TUNEL method.Results:The injury area in mice brain and gastric mucosa was significantly different between group B and group A, C(P<0.05).The content of HSP70 protein markedly higher on the brain and gastric mucosa after model was induced,respectively.Group B was notably higher than group A(P<0.05).Group B,D was observed apoptotic cells obviously, which was different from group A,C,significantly.GGA pretreatment enhanced the up-regulated expression of HSP70 and decreased the apoptotic index distinctly(group C vs. group B,D;P<0.05).Conclusions:The improved Feeney impact model can lead to TBI related AGMLs on mice.The levels of HSP70 in mice brain and gastric mucosa were significantly high following TBI.Cells apoptosis was one of the characteristic TBI related AGMLs.GGA can induce HSP70 protein expressed in mice brain and gastric mucosa.HSP70 was concedned with the process of inhibition apoptosis.GGA can be uesed as prevention and therapy in TBI related AGMLs.
