Objective: Resveratrol is one kind of polyphenol compounds derived from plant extracts. There have been many studies on the relation between resveratrol and cancer including almost all types of cancer. Many studies were found that resveratrol has a wide range of activities including anti-proliferation, metastasis inhibition, anti-apoptosis and so on in many carcinoma cells. However, the effects of Resveratrol on human tongue squamous carcinoma Tca8113 cells are not reported until now. Therefore, in this study we used Tca8113 cells to investigate the inhibitory effects of resveratrol on the metastasis of Tca8113 cells, and to explore its molecular mechanisms.Methods: In this study,the effects of resveratrol on Tca8113 cell proliferation were done using sulforhodamine B(SRB) assay. Cell apoptosis was determined by DAPI nuclear dying assay. Transwell chamber assay was adopted to study the inhibitory effects of resveratrol on the migration and invasion of Tca8113 cells. Cell adhesion was done by cell-matrix adhesion assay. The MMP-2 and MMP-9 expressions in Tca8113 cells were assessed through Gelatin Zymography assay. The effect of resveratrol on NF-κB nuclear transfer capacity was conducted by immunohistochemistry assay. The protein expressions of Akt, and p-Akt, and IKK and p-IKK were done using Western blotting analysis.Results: Resveratrol inhibited the proliferation of Tca8113 cells in a time- and concentration-dependent manner. The lower concentrationsof resveratrol(<18.5 μg/m L) did not induce cell apoptosis and had growth retardation, but its concentrations(>18.5 μg/m L) could induce cell apoptosis. Resveratrol dependently inhibited the invasion, migration, and adhesion viabilities of Tca8113 cells. Moreover, the expressions of MMP-2 and MMP-9 were reduced in a concentration-dependent manner. Immunohistochemical assay showed that with the increase of the concentration of resveratrol, the capacity of p65 entering into the nucleus gradually reduced. The expression of Akt had no a significant change, but the expressions of p-Akt, IKK, p-IKK were reduced in a concentration-dependent manner.Conclusion: Resveratrol can inhibit the proliferation, invasion, migration, adhesion and expressions of MMP-2 and MMP-9 in Tca8113 cells. This suggests that resveratrol can inhibit the metastasis of Tca8113 cells. Its mechanism maybe attribute to PI3K/Akt and NF-κB signaling pathway.
