Objective To study the effect of heat shock protein 72(HSP72) on apoptosis of rat glomerulurs endothelial cells(r GENCs) in hypoxic environment. Methods Cobalt chloride(CoCl2)was used to provide hypoxia circumstance in vitro.The rGENCs were exposure to hypoxia which induced by CoCl2 at different treatment time periods(24h, 48 h, 72h), the control group was exposed to normal environment.The cell apoptosis were detected by flow cytometry.Then,the rGENCs were divided into 5 groups(control group, hypoxia group, hypoxia+DMSO group, hypoxia+HSP72 inhibitor group, hypoxia+HSP72 agonist group) according to the different intervention methods. The cell apoptosis and content of HSP72 were detected by flow cytometry and westernblot respectively. Results With the elongation of hypoxia time, the apoptosis of the cells increased gradually, and reached the upper point at 72 h, there was a statistically significant difference compared with the control group(23.80 ±0.75% vs 2.4±0.2%,P<0.01).The expression of HSP72 in rGENCs was increased significantly under hypoxic conditions compared with that in control group(0.82±0.15 vs 0.23±0.09,P<0.01). When the content of HSP72 was decreased caused by co-culture with HSP72 inhibitor, a further increase of apoptosis of cells had been found. However, the cells apoptosis was decreased when the content of HSP72 increasion caused by co-culture with HSP72 agonist. Conclusion Hypoxia can induce apoptosis increasing of rGENCs, HSP72 is an important factor which affect the apoptosis of rGENCs under hypoxia circumstance, and it’s content has a negative correlation with the apoptosis of rGENCs.
