Objectives To investigate the regulation of acupuncture serum on the expression of p38 MAPK in asthmatic rats airway smooth muscle cells(ASMCs), and explore its action mechanism.Methods We employed rats as the model organism, challenging them with OVA to set up the Asthma model. Groups were given accordingly interventions: acupuncture group treated by acupuncture from the day rats get OVA for aerosol challenge, rats in model group challenged with OVA without treated and blank group only get saline. Taking the lung compliance and lung elastic resistance at the end of aerosol challenge. Then the rats of the acupuncture group and the blank group were fixed when they are anesthetized to take blood from aorta abdominalis, standing for an hour. Centrifuging the blood to get the liquid supernatant, and then get bacteria killed and inactivated. In addition, the ASMCs were primarily cultured via the method of enzymatic digestion without sieve filtration, and identified by the assays of morphological and immunocytochemical. Furthermore, we used acupuncture serum to achieve the intervention for rats ASMCs, and the concentration in ASMCs of asthmatic rats were detected by MTT. Finally, we detected the expression level of p38 MAPK through Immunocytochemical method, Western blot and RT-PCR.Results 1 Compared with the control group, the rats in model group are decreased with lung compliance and increased with lung elastic resistance; Compared with the asthmatic model group, rats in acupuncture group are increased with lung compliance and decreased with lung elastic resistance. 2 Compared with the control group, the inflammatory cells in the BALF of rats in model group increased significantly; Compared with the model group,the inflammatory cells in the BALF of rats in the acupuncture group decreased. 3 HE staining of the rats lung tissue showed that, rat bronchial wall in model group has the massive inflammatory infiltration, mucus in the lumen and increased red blood cells.Besides, the asthmatic airway walls are thicken and deformated; Compared with the model group, there is less inflammatory cells, mucus and red blood cells in the lung tissue of rats in acupuncture group. 4 The method of enzymatic digestion without sieve filtration can obtain high purity and a large number of airway smooth muscle cells. 5 20% of the acupuncture serum for asthmatic rats ASMCs have the greatest impact on the proliferation ability. 6 The expression of p-p38 MAPK in M+C group(blank serum intervention in asthmatic rat airway smooth muscle cells) is significantly higher than that of the cells in the control group and the asthma model group with acupuncture serum and blocker intervention. 7 The gene expression level of p38 MAPK in group of blank serum intervention in asthmatic rat airway smooth muscle cells is higher than that of the cells in the control group and the asthma model group with acupuncture serum and blocker intervention.Conclusions 1 The aerosol challenge with OVA can lead to the occurrence of respiratory dysfunction and airway inflammation for rats, and acupuncture can improve the symptoms of asthma. 2 20% of the acupuncture serum for asthmatic rats ASMCs is the optimal concentration. 3 Acupuncture serum is useful for asthma by decrease the expression of p38 MAPK in asthmatic rats airway smooth muscle cells.
