Objective Analyzing the expressions of histone H4 acetylated and histone deacetylase(HDAC) in embryo lemma was aimed to reveal the relationship between the acetylated histone and premature rupture of membranes(PROM), and to provide the basis for PROM prevention.Methods 1 Object of study: Selecting the natural birthing mother with singleton as objects, and who were treated in Tangshan Maternal and Child Health Hospital during the same period. According to the occurrence of premature rupture of membranes and break the membrane, they were divided into three groups, which are preterm rupture of membrane delivery group(Full-term labor, FTL), term premature rupture of the early broken delivery group(the term premature rupture of membranes, TPROM) and not term premature rupture of the premature rupture of delivery group(preterm premature rupture of membranes, PPROM), 30 cases in each group. 2 Population survey: Recording objects’ general information like age, education, occupation, body mass index(BMI),tobacco use and alcohol use and so on. Collecting the objects’ laboratory indexes like blood zinc, blood calcium, blood glucose, total cholesterol, blood albumin, neutrophilic granulocyte percentage, eosinophils percentage and so on. Collecting the organization of membranes tissue and stored at-40℃.3 Laboratory testing: Testing the expression of H4 acetylation in organization of membranes via Western blot; testing the expression of histone acetylation enzyme, HDAC-2, HDAC-1, HDAC-6 of the expression of matrix metalloproteinase MMP-2, MMP-8, MMP-9 expression of inflammatory cytokines IL-1β,IL-6, TNF-α expression and cellular apoptosis Caspase-3, Caspase-8 expression of.4Statistical analysis: the object of study in general, all test indicators, measurement data with mean±standard deviation(—x + s), said the count data to chi-square(chi square),according to the indexes for normality and f test, the effect of different factors analysis by using single factor analysis of variance and SNK test between groups, multiple factors analysis using covariance analysis model, all the statistical analysis using bilateral inspection, inspection level of α= 0.05.Results 1 General situation analysis: three groups of age, race, occupation, education level, and tobacco and alcohol addiction, maternal inferior generally balanced comparable, BMI high level may promote the occurrence of premature rupture of membranes. The laboratory indicators related with premature rupture of membranes 2single factor analysis: 1) low blood zinc status may promote the occurrence of premature rupture of membranes before term, neutrophil percentage increased, low blood calcium and low albumin levels associated with the occurrence of premature rupture of membranes periods(P<0.05), and other laboratory examination index there was no statistically significant difference in the distribution of the three groups(P>0.05). 2)MMP-2 and MMP-9 expression status of matrix metalloproteinase exhibits increased trend as TPROM <FTL < PPROM group(P <0.01), MMP-8 expression had a significant effect(P <0.01) for PPROM occurred, and its term delivery and whether the rupture seems no correlation(P>0.05). 3) Inflammation related factor IL-6 and TNF-α rise in expression is associated with the occurrence of PPROM, but has no obvious relation with the incidence of TPROM, IL-1 beta lower expression is related to whether the membranes rupture(P<0.01) and may has nothing to do with the broken membrane gestational age(P>0.05). 4) apoptosis factor Caspase 3 in the group of membranes ruptured and unruptured comparison were high expression(P<0.01), rupture of no difference between two groups(P>0.05), prompted the activation of Caspase-3 only related to the rupture of membranes and has nothing to do with whether not term premature rupture of membrane; Caspase-8 in three groups were expressed in fetal membrane organization but not statistically to compare the difference(P>0.05). 3Histone H4 acetylation and acetylation enzyme, HDAC-2, HDAC-1, HDAC-6 of expressing the relationship between premature rupture of membranes and analysis: 1) the present H4K5 in premature rupture of membranes in the organization, high H4K8 acetylation state of expression, and FTL <TPROM<PPROM group gradually rise, the significant difference(P<0.001), suggesting that the histone H4 acetylation to participate in the occurrence of premature rupture of membranes, premature rupture of membranes and promoted the early. 2) HDAC-1, HDAC-2 and HDAC-6 positive inhibition were premature rupture of membranes in group(P<0.001), inhibit the most significant PPROM group, that group of proteins to inhibit the acetylation enzyme can lead to premature rupture of membranes occurred earlier. 4 of the various factors associated with premature rupture of membranes, according to the results of analysis of covariance histone H4K5,H4K8 acetylated expression enhancement to promote all the occurrence of premature rupture of membranes gestational age(P<0.05) or less, histone acetylation enzyme HDAC-1, inhibition of HDAC-2 also significantly promote the occurrence of premature rupture of membranes(P<0.05), a histone to acetylation enzyme HDAC-6 changes on does not affect the occurrence of premature rupture of membranes(P>0.05).Conclusion 1 High expression of histone acetylated may be involved in the occurrence of premature rupture of membranes. 2 The expression of fetal membranes of histone H4 acetylation enhancement and histone deacetylase HDAC-1 and HDAC-2 inhibition significantly promote the premature rupture of membranes(PROM) occurs.
