Objectives To investigate the effect and action mechanism of Yiqi Yangyin Huoxue Decoction(YYHD) improve insulin resistance in type 2 diabetes rats by measuring the expression level of mi RNA-126 in liver.Methods Sixty 12-weeks-old wistar rats, after the adaptive feeding one week, were randomly divided into blank group(n=10) and diabetic group(n=50). Type 2 diabetic rats model was induced by feeding high fat diet combined with intraperitoneal injection of a small dose of streptozotocin(STZ) in diabetic group rats. The successful type 2diabetic rats into the testing group were randomly divided into 5 groups: model group,Jinlida group, low dose of YYHD group, middle dose of YYHD group and high dose of YYHD group. Normal saline, Jinlida, different doses of YYHD(low, middle, high) were used to intervene type 2 diabetic rats, and normal saline was used to interene blank group.During the intervention process, weight and FBG were measured weekly. After 5 weeks of intervention, these rats were fasted 12 hours after the last dose, then draw blood from abdominal aorta to detect fasting insulin(FINS), blood lipid(TC, TG, LDL-C, HDL-C)and other biochemical indicators, calculate Insulin sensitivity index(ISI) and Insulin resistance index(HOMA-IR). Subsequently these rats were all executed and their liver tissues were harvested. The morphological changes in liver were observed by HE staining.The expression level of mi RNA-126 was detected in rat liver tissue by RT-PCR technique, for investigating action mechanism of YYHD improve hepatic insulin resistance in type 2 diabetes rats.Results 1 Building model of type 2 diabetes rats: Type 2 diabetic rats model was induced by feeding high fat diet 4 weeks combined with intravenous injection of a small dose of streptozotocin(STZ) in Wister rats. With FBG≥16.7mmol/L for successful model.2 Effect on weight of rats in each group: middle dose group and high dose group of YYHD and Jinlida decreased weight in type 2 diabetic rats significantly(P<0.05). But there were no statistical differences between the three(P > 0.05). 3 Effects on FBG,2h BG, FINS, ISI and HOMA-IR of rats in each group: Compared with model group,different dose group of YYHD and Jinlida group could reduce the level of FBG, 2h BG,FINS, HOMA-IR, and improve the level of ISI to different extents. The difference was statistically significant(P < 0.05). Middle dose group of YYHD is the best in these groups(P<0.05). 4 Effect on blood lipid of rats in each group: Compared with model group, the level of TC, TG, LDL-C could reduce of different dose group of YYHD and Jinlida group to different extents, the level of HDL-C has increased. the difference was statistically significant(P<0.05). Middle dose group of YYHD is the most obvious in terms of decreasing TC and LDL-C, Which is better than Jinlida group(P < 0.05). 5Effect on liver morphology of rats in each group: In blank group, the liver tissue is normal, clear and complete, no obvious pathological changes, no hyperplasia, edema,fatty degeneration and so on. In model group, the damage of liver tissue is obvious, the hepatic cords arrange in disorder, with swelling and degeneration. There are patchy necrosis in the small leaf, and a large number of inflammatory cells infiltration.Compared with model group, different dose group of YYHD and Jinlida group can alleviate the pathological damage of rat liver to some extent, especially middle dose group of YYHD is most obvious, showing that liver cells are normal basically, and a small amount of fatty degeneration cells and inflammatory cells are seen occasionally.The liver tissue damage slightly. 6 Effect on the expression level of mi RNA-126 in liver tissue of rats:compared with model group, the expression level of mi RNA-126 reduce to different degrees in different dose group of YYHD and Jinlida group(P < 0.05);Compared with Jinlida group, middle dose group of YYHD decrease mi RNA-126 expression level more obviously, the difference has statistical significance(P<0.05).Conclusions YYHD can improve insulin resistance of type 2 diabetic rats and has a protective effect on the damaged liver. The mechanism may be reducing the expression level of mi RNA-126 in the liver.
