BackgroundBasic research shows that low intensity ultrasound irradiation hyperplasia of the breast tissue, can induce apoptosis of mammary gland hyperplasia tissues; Clinical research shows that low intensity focused ultrasound treatment of mammary gland hyperplasia can block the myelinated nerve fibers and relieve pain, and can keep for a long time.We Can assume that low intensity ultrasound treatment of mammary gland hyperplasia may also exist can long time effect of control factors, hormone receptor levels in breast tissue may be such factors. So we adopt the low intensity ultrasound irradiation experimental hyperplasia of mammary glands in New Zealand white rabbit, to discuss low intensity ultrasound on treating hyperplasia of mammary gland hormone receptor. ObjectiveEstablish animal model of hyperplasia of mammary glands to observe the effects of Low-intensity ultrasound on expression of hormone receptors in rabbit model of mammary gland hyperplasia.Methods1. 16 female Newzealand rabbits were randomized into 2 groups:A,B, N = 8, A is the control group, B for the model group. After modeling according to the change of nipple height and pathological biopsy results to determine whether a model success.2. 24 female Newzealand rabbits were randomized into 3 groups:A,B and C, N = 8, Group A(control group)will not be modeling and treatment,Group B(sham treatment group) and group C(treatment group)modeling;after modeling,Group B were treated with sham treatment,Group C to give low-intensity ultrasound irradiation.Pathological changes and the expressions of ERα,ERβand PR were measured. Results1.After modeled, the height of breast nipple in group B is(6.28±0.17)mm, compared with group A significantly higher, there are statistically significant(P<0.001), by observation of pathological tissue section, the number of breast lobular,alveolar and ductal are increased, interstitial fibers increased significantly, reach the standard animal model of hyperplasia of mammary glands.2.(1)After modeled, the height of breast nipple in B and C respectively are(6.11 ± 0.14)mm,(6.10 ± 0.13)mm, compared with group A were significantly increased, These differences have statistical signifficance(P<0.05), B and C are no statistical significance between two groups(P>0.05), it test that Using the same method to establish the animal model of consistent results. After treatment, the height of breast nipple in group A is(1.93±0.07)mm, the height of breast nipple in group B is(6.13±0.16)mm, the height of breast nipple in group C is(3.36±0.14)mm, two-two comparisons among the means were done by LSD method, Group B nipple height is still higher than group A, there are differences between the statistical(P<0.05),it test that hyperplasia of mammary gland organization does not offer any intervention in the treatment of cases, hyperplasia of mammary glands won’t recover, group C compared with A and B have statistical significance(P<0.05),after be treated, group C nipple height decrease obviously, but has yet to return to normal levels.(2) The number of breast lobular,alveolar and ductal in group B respectively are 8.37±1.71,10.25±1.34,5.31±1.19, they are compared with group A were significantly increased(P<0.05), accord with standard of hyperplasia of mammary glands;after be treated, the number of breast lobular,alveolar and ductal in group B respectively are 4.37±1.82,6.93±1.98,3.87±0.96, compared with group B were statistically significant(P<0.05).(3)The expression of ERα and PR respectively are 4.77 ± 2.23,6.18 ±2.22,compared with group A were significantly increased(P<0.05), ERα and PR with hyperplasia of the breast tissue are the same; after treated, the expression of ERα and PR decreased 3.24±2.44, 3.25±1.34, compared with group B were statistically significant(P<0.05), it tested that low intensity focused ultrasound can be cut hyperplasia of mammary gland tissue expression of hormone receptors. The expression of ERβ in three groups are 3.82 ± 2.66, 3.85 ± 2.65, 3.96 ± 2.54, comparison between groups was no statistical significance(P>0.05).ConclusionsRegulation of estrogen and progesterone receptor expression is an important mechanism of low-intensity focus ultrasound breast hyperplasia treatment.
