MCF-7 cells were transfected with plasmid pc DNA3.1/WNT5 Band pcDNA3.1) respectively, and the expression of WNT5 B mRNA and protein level were examined in two group by Real-time PCR and Western blotting. Subsequently, the changes of cell proliferation and cell apoptosis were detected by CCK-8 assay and Flow Cytometry respectively. Results:The expression of WNT5 B in breast cancer lines were lower than in MCF10 A cells. The WNT5 B expression in MCF-7 cells in experimental group was significantly higher than that in vector group(P<0.05). However, the abilities of cell proliferation in experimental group decreased significantly as compared with those in vector group(P < 0. 05).The number of cells in S-phase obviously increased while the percentage of cells in G1-phase and G2/M-phase decreased compared with vector group. The number of apoptosis cells in WNT5 B group was significantly increased than that in vector group. Conclusion : The expression of WNT5 B was decreased in breast cancer cells. WNT5 B overexpression significantly inhibited the proliferation and promoted cell apoptosis of breast cancer cell line MCF7. | Keywords/Search Tags: | WNT5B, MCF-7, proliferation, apoptosis | PDF Full Text Request | Related items |
