Objective: To measure and compare the vulvar tissue thickness of patients with NNEDV and normal patients, provide the date for NNEDV diagnosis.Methods: 263 slices of vulvar tissue(248 with NNEDV and 15 with normal vulvar tissues) in the second affiliated hospital of chongqing medical university from January 2013 to June 2015 were collected. 202 NNEDV slices meet the study requirements, which included 51 cases of vulvar squamous cell hyperplasia,43 cases of vulvar sclerosing lichen and 108 cases of mixed type NNEDV. The slice was observed by bright field microscope and the vulvar skin thickness was measured by ISCapture imaging analysis software and the data was analysised by SPSS20.0 statistical analysis software.Results:(1) the thinkness of epidermis and dermis of normal vulvar skin was(90.47±16.84)μm and(460.53±107.25)μm respectively. The stratum corneum and stratum granulosum of vulvar skin was significantly thicker in the patients with hyperplasia NNEDV than normal vulvar skin and patients with atrophy NNEDV and mixed type of NNEDV(P < 0.01), The stratum spinosum and stratum basale of vulvar skin was significantly lower in the patients with atrophy NNEDV than normal vulvar skin and patients with hyperplasia NNEDV and mixed type of NNEDV(P < 0.05). There was no difference in the epidermal thickness between normal vulvar skin and patients with NNEDV(P > 0.05).(2) The dermal papilla layer of vulvar skin was significantly thicker in the patients with hyperplasia NNEDV than normal vulvar skin and patients with atrophy NNEDV and mixed type of NNEDV(P < 0.05), The dermal papilla layer of vulvar skin was significantly lower in the patients with atrophy NNEDV than normal vulvar skin and patients with hyperplasia NNEDV and mixed type of NNEDV(P > 0.05). There was no difference in the thickness of reticular layer between normal vulvar skin and patients with NNEDV(P > 0.05).Conclusion: The thickness of stratum corneum, granulosum and dermal papilla layer was increased in patients with hyperplasia NNEDV and the thickness of stratum spinosum and basal layer and dermal papilla layer was decreased in patients with atrophy NNEDV. Which is useful for vulvar skin diagnosis model establishment.
