Objective:To analyze clinical and biological characterisis and progonis of AML1/ETO positive childhood acute myeloid leukemia(AML).Methods:Retrospectively studied the 33 newly diagnosed acute myeloid leukemia children with AML1/ETO,who were enrolled in the study from January 2011 to May 2015.Conclusion the characteristics in morphology,immunology and molecular and the probabitity of event free survival was estimated by Kaplan-Meier analysis.Result:1.Of all the 33 AML1/ETO positive children,the ratio of male and female was 1.54:1;the median age was 7 years and 3 months;20 children the onset WBC>10×109/L.2.MICM types:(1)Morphology:M2 87.9%(n=29),M5 6.1%(n=2),M4 3%(n=1),MDS-AML 3%(n=1);(2)Immunology:25 children’s immunology were AML,8 were Ly+AML;1 child the CD19 was positive;2 the CD56 was positive.(3)Cytogenetic:All the children who have this test were examed to t(8;21)(q22,q22),23 of the children have additional abnormal chromosome,mainly lack of sex chromosome.5children have complex karyotype.3.In this study,the complete remission(CR)rate of induction chemotherapy is 67%.30 cases complete the induction chemotherapy and consolidation chemotherapy.The CR rate after two courses is 80%.No Treatment Related Mortality of children.4.The median follow-up time of 25 children who were in the follow-up is 18(8~59)months.The median overall survival time was 14 months and median event free survival time was 9 months. The median relapse time of 5 relapsing children was 10 months;There was no patient relapsed within 6 months after CR.5.Univariate analysis indicated that complex karyotype was independent unfavourable factor which has lower overall survival rate and event free survival rate.Conclusion:AML1/ETO-positive AML children had a higher complete remission rate,but there was not significant improve of long-term prognosis.The karyotype of chromosome is one of the factors affecting the long-term therapeutic effect.
