Objective To explore the expression levels of m RNA and protein about eukaryotic translation elongation factor 1 alpha 2(eEF1A2) gene and PI3K-Akt signaling pathway of genes in cervical cancer tissues. To clarity of eEF1A2、PI3K, Akt and HPV16L1 genes expression related to cervical cancer tumorigenesis and development. This research will lay the foundation for prevention and treatment of cervical cancer.Methods 1. Collected 72 cases of cervical cancer, 46 cases of cervical intraepithelial neoplasia and 40 cases of chronic cervicitis of paraffin tissues for immunohistochemistry experiment. Collected fresh cervical cancer tissues and normal cervical tissues for q RT-PCR experiments. Culture He La and Si Ha cervical cancer of cell lines and human umbilical vein endothelial cells were used for Western blot experiments. 2. Select the β-actin gene as an internal control, using real-time quantitative reverse transcription-polymerase chain reaction(q RT-PCR) method for detection of m RNA expression levels of eEF1A2, PI3 K, Akt gene. 3.eEF1A2, PI3 K, p-Akt and HPV16 L1 protein expression levels in cervical cancer, CIN and chronic cervicitis tissues were detected by Immunohistochemical SP method. 4. eEF1A2,PI3 K, p-Akt protein expression levels in He La, Si Ha and human umbilical vein endothelial cells were detected by Western blot method. SPSS17 software was used in all statistical analyses. Results were considered statistically significant if P < 0.05.Results 1. qRT-PCR results showed that the level of mRNA expression between eEF1A2,PI3 K, Akt gene in cervical tissue were higher than that in normal cervical tissue. 2.Immunohistochemistry results showed the eEF1A2, PI3 K, p-Akt protein expression level in cervical cancer tissues were higher than that in cervical chronic cervicitis tissues, and HPV16 L1 protein expression level in cervical chronic cervicitis tissues were higher than that in cervical cancer tissues. The differences are statistical significant(P <0.005). 3. The expression of eEF1A2, PI3 K, p-Akt protein in He La and Si Ha of cervical cell lines were higher than that of normal epithelial cells.Conclusions HPV16 L1 protein expression level is beginning to reduce in CINIII and protein expression level reduce more and more in invasion of cervical cancer, it may be use for diagnosis of CINII and CINIII or CINII and cervical cancer. The results suggest that aberrant expression of eEF1A2, PI3 K and AKT genes may play a certain role in the tumorigenesis and development of cervical cancer.
