OBJECTIVE:To observe the effect of different segments of the renin-angiotention-aldosterone system on renal tissue in rats with unilateral ureteral obstruction.METHODS:Fifty SD rats aged 6 weeks were randomized into five groups, which were normal group, model group, enalapril group, valsartan group and apironolactone group,with 10 rats in each. The UUO models were induced by ligating the left ureter. In the normal group and model group, the rats received normal saline 10ml/kg everyday with intragastric instillation from 24 hours before the obstruction to the 2 weeks after the obstruction. The treatment groups received intragastric instillation daily with enalapril10mg/kg, valsartan 10mg/kg, apironolactone 100mg/kg from 24 hours before the obstruction to the 2 weeks after the obstruction. Each group was sacrificed at 2 weeks after UUO was initiated. Collecting the sample of blood and urine to biochemistry detection. The renal tissue was evaluated by Hematoxylin—eosin staining and observed the expression of TGF-β1 m RNA, AGT m RNA, ACE m RNA, ALD m RNA by RT-PCR.RESULTS:The levels of Scr and BUN were significantly increased in model group,enalapril group, valsartan group and apironolactone group compared with those in normal group(all P<0.05). The levels of Scr and BUN were increased in model group compared with those in the three treatments groups( P< 0.05).And there were no significant differences among the three treatment groups. There were no significant differences at 24 h urine protein among five groups(P>0.05).CONCLUSION:The blockage of renin-angiotention-aldosterone system at different sites have the same inhibition of renal interstitial fibrosis, and the result is similar.
