In female malignant tumors, cervical carcinoma is one of the most common malignancies. Radix scutellariae is a kind of natural herbs and one of the commonly used Chinese medicinal herbs in our country. Meanwhile Scutellaria baicalensis Georgi has extensive pharmacological action and value on clinical application. Other studies have shown that radix scutellariae has effect on antitumor. At present, the combination of baicalin and baicalein haven’t been reported in cervical cancer treatment. In this paper, in vivo experiments have been used to study the function and mechanism of the combination of baicalin and baicalein against cervical cancer.In this research, the combination of baicalein and baicalin was used on the U14 cervical cancer mouse model to detect tumor inhibitory rate of solid tumor model and life prolonging rate of ascitic fluids model. In order to study the effect and mechanism of the combination of baicalein and baicalin on cervical cancer, H.E staining technique has been used to observe the the cell morphology of cervical-cancer cells, flow cytometry has been used to observe cell cycle distribution and the rate of tumor cell apoptosis. And we also used immunohistochemical technique to detect the expression of apoptosis-associated protein. In addition, in order to research the effect of the compound on the immune system, spleen index and thymus index were counted, the content of TGF- beta 1 and IL-10 were detected by enzyme linked immunosorbent assay(ELISA). The results suggested that, the combination of baicalein and baicalin could inhibit the growth of tumor cells and prolong survival time of ascites tumor model compared with negative control group. Its possible mechanisms were blocking the cell cycle of tumor cells at G0/G1 phase, adjusting the expression of gene cytochrome C, caspase-9 and caspase-3, and further inducing U14 tumor cell apoptosis. Besides, the combination of baicalein and baicalin could increase spleen and thymus index and decrease the content of TGF-β1 and IL-10, thus to enhance the immune function of tumor-bearing mice and improve the anti-tumor ability of mice.This research was based on the U14 cervical cancer mice model and mainly deals with two aspects, inducing tumor cell apoptosis and improving the organism immunity. This study initially investigated the mechanism of the compound against cervical cancer and laies the solid foundation for the further research on anti-cancer effect of Scutellaria baicalensis Georgi.
