Objective: Pulmonary malignant tumor has high disability rate and case-fatality rate,the traditional chemical treatment is usually over treatment difference resistant, appear adverse reaction after chemotherapy, will seek to improve drug cisplatin sensitivity, thus to reduce the use of cisplatin dose so as to reduce side reaction of cisplatin will be more important in clinical value.This study was to evaluate the coordination of parthenolide with cisplatin on lung adenocarcinoma A549 cells from the level of molecular biology. Whether PTL can increase the sensitivity of lung adenocarcinoma A549 cells to cisplatin, to evaluate the synergistic sensitization effect of the coordination of parthenolide with cisplatin on lung cancer A549 cells.Methods: The subjects is the logarithmic growth of A549 cells.After treated by parthenolide and(or) cisplatin, the viability of cell lines was detected through microscope and MTT. According to the value of q so as to know whether the coordination of parthenolide with cisplatin has a synergy on A549 cells. The apoptosis of cell lines and cell cycle was detected by FCM. The expressions of apoptosis inhibiting gene Bcl-2 and apoptosis gene Bax mRNAs were measured by real-time-PCR.Results: Determined by MTT/FCM/PCR,the results showed that small doses of parthenolide have an effect on promoting proliferation of A549 cells, high-dose of parthenolide can inhibit proliferation of A549 cells.The apoptotic rate was significantly increased in A549 cells treated with PTL in combination with cisplatin, compared with that in cisplatin-treated or PTL-treated A549 cells, and the cell cycle was arrested at phase G1+S.The expression level of Bcl-2 protein was decreased(P<0.05) and the expression level of Bax was increased(P < 0.05) in a concentration-dependent manner in A549 cells treated with different, and these effects were significantly enhanced in A549 cells treated with PTL in combination with cisplatin as compared with those in the cells treated with cisplatin or PTL alone(P<0.05).Conclusion: In vitro, high-dose of PTL can inhibit the proliferation of human lung adenocarcinoma A549 cells and induce its apoptosis, what is more, the coordination of parthenolide with cisplatin have a synergistic efect on inhibiting cellular proliferation and inducing cellular apoptosis ofc lung adenocarcinoma A549 cells,which may probably be associated with the increased proportion of G1+S phase cells, to induce apoptosis,with the decrease in the ratio of Bcl-2/Bax mRNAs.
