Traditional Chinese medicine(TCM) adulteration incident happened frequently, the rapid on-site detection of TCM adulteration has been widely concerned. As a matter of fact, there are many kinds of TCM composition is complex, the quality standard is not perfect. The study is to establish TLC-SERS methods for TCM illegally added with chemical drugs. The research object is more than ten kinds of chemicals which illegally added in the same type of TCM(lipid-lowering and heat-clearing TCM), including the adulteration of common adulterated drug and the possible components in the future. Providing new ideas and methods for the determination of chemicals which illegally added in the other type of TCM.This study was carried out in the following two aspects of work:The first part is about the detection of eleven kinds of chemicals easily adulterated in lipid-lowering TCM. The eleven chemicals are nicotinic acid, lovastatin, simvastatin, probucol, fenofibrate, bezafibrate, ciprofibrate, atorvastatin calcium, rosuvastatin calcium, fluvastatin sodium, and pravastatin four methyl amine. The experiment gives a detailed work about the TLC separation and SERS detection. By investigating the influence of various experimental parameters, including the type of TLC plate, mobile phase, silver sol preparation method, coagulating agent, pH value, the silver sol dropping amount, laser power, detection limit and so on, as well as optimizing TLC and SERS detection conditions, the initially established TLC-SERS method was used to detect positive samples and real TCM samples. The detection results of real samples has been verified by UPLC-QTOF/MS.The second part is about the detection of four kinds of chemicals easily adulterated in heat-clearing TCM. The four chemicals are aminopyrine, indomethacin, naproxen and ibuprofen. The experiment by investigating the separation conditions of thin layer chromatography and detection conditions of SERS. TLC-SERS method was preliminary established for rapid detection of four kinds of chemicals added illegally in heat-clearing TCM. Later work will be on the other seven adulteration of medicine(namely aspirin, acetaminophen, salicylic acid, butazodine, piroxicam, diclofenac sodium, clobetasol propionate), which were regarded the research object, and then establish the method of TLC-SERS, in order to achieve the purpose of the simultaneous detection of eleven kinds of drugs in heat-clearing TCM.This study focused on the rapid detection of adulteration of TCM field and the results are as follows:1.The detection results of eleven kinds of chemicals easily adulterated in lipid-lowering TCM: Study selected type of thin layer plates are Merck plate GF254; The mobile phase is petroleum ether-ethyl acetate-acetic acid(5.5:2.8:1), which can achieve the purpose of eliminating effect of matrix and separating of eleven kinds of chemicals; substrate is silver sol using DMF as solvent; The silver sol dropping amount is 5uL; The laser power is 100%; The limit of detection of chemicals are below 1%, the limit of detection are as follows: nicotinic acid 0.05%, lovastatin 0.1%, simvastatin 0.1%, probucol 0.01%, fenofibrate 0.01%, bezafibrate 0.002%, ciprofibrate 0.01%, atorvastatin calcium 0.005%, rosuvastatin calcium 0.01%, fluvastatin sodium 0.02%, and pravastatin four methyl amine 0.02%.2.The detection results of four kinds of chemicals easily adulterated in heat-clearing TCM: The Merck plate GF254 is selected; The mobile phase is ethyl acetate-methenyl trichloride-acetic acid(10:10:0.3); Substrate is silver sol using DMF as solvent; The silver sol dropping amount is 5uL; The integration time is 15s; The limit of detection are as follows: aminopyrine 0.02 mg/mL, indomethacin 0.2 mg/mL, naproxen 0.1 mg/mL and ibuprofen 0.02 mg/mL. The rest of the seven chemicals remains to be studied.This study established a method of TLC-SERS for the rapid detection of several chemicals in lipid-lowering(or heat clearing TCM) at same time, and successfully detected the chemicals in the real TCM sample. With the advantage of rapidity, simplicity, high sensitivity and specificity, the method has good application prospect in the rapid detection of illegal adulteration on TCM, and provides the experimental basis for the TLC-SERS machine which is being researched and developed.
