Background Lung cancer is the most common human malignant tumor in the world,with the highest morbidity and mortality. Non-small cell lung cancer(NSCLC) accounts for approximately 85% of all lung cancer cases.More than 90% of patients cause of death were associated with tumor metastasis.A better understanding of the molecular mechanisms underlying lung cancer formation and progression should be helpful in developing more effective treatments for this disease. A variety of different cancers has been reported overexpressed TRIM29, but the exact function of TRIM29 in cancers is not fully understood.In 2014,Ai et al found that TRIM29 shows close relationship between Twist1 in breast cancer research,which suggested the mechanism of tumor metastasis. To clear explain TRIM29 biological characteristics in human non-small cell lung cancer cells, this study selected human non-small cell lung cancer A549 cell as a specimens, and investigate the effect of silencing TRIM29 on the invasion and the influence of associated proteins Twist1 and E-cadherin expression in human non-small cell lung cancer cells.Objective The aim of this study was to explore the effect and mechanism of TRIM29 on migration of human non small cell lung cancer.Methods1.RNA interference and Virus mediated methods were employed to establish the stable transfected A549 cells with TRIM29 sh RNA.2. Real time reverse transcriptase polymerase chain reaction and Western blotting assay were used to detect TRIM29 m RNA and protein expressions in A549 cells which transformed with TRIM29 sh RNA.3.Transwell invasion assay was employed to determine the cell migration of A549 cells which transformed with TRIM29 sh RNA.4.Western blotting assay was used to detect Twist1 protein and E-cadherin protein expressions in A549 cells which transformed with TRIM29 sh RNA.Results1. We transfected A549 cells with TRIM29 specific small interfering RNA(si RNA) vector and the negative control cell lines with empty vector transfection. Compared with untreated group or the si CONTROL group, the m RNA levels of TRIM29 in A549 cells with TRIM29 sh RNA showed no significant between them,however, the protein expressional levels showed significantly decreased.2. We studied the effect of TRIM29 knockdown on the invasion of A549 cells using the transwell invasion assay. The results showed that invasion was significantly inhibited in the TRIM29 knockdown cells compared with the untreated group, or the si CONTROL group.3.Western-blotting further studied the protein expression of Twist1 and E-cadherin. The results showed that after suppressing the expression of TRIM29, the expression of Twist1 was down-regulated and the expression of E-cadherin was up-regulated.ConclusionOur study was aim to explore the effect and mechanism of TRIM29 on biological behavior of human non small cell lung cancer. The results showed that after suppressing the expression of TRIM29, the invasion of A549 cells was significantly inhibited.The mechanism of TRIM29 on biological behavior of human non-small cell lung cancer may by regulatting the expression of Twist1 and E-cadherin.It our findings suggest that TRIM29 may be a new therapeutic application in treating human lung cancer.
