Objective:To establish model of zebrafish vascular inhibition and cardiac injury and methods of drug screening and investigation; observe indicators of zebrafish embryonic vascular and heart; study inhibition effects of kayaku oxaliplatin, 5-fluorouracil and Chinese medicine compound kushen injection when used alone, in combination and together; study restoration and protection effects of resveratrol and danhong injection on cardiac injury caused by astemizole and analyze influence of these medicine on zebrafish embryonic vascular and heart.Methods:Utilize transgenic zebrafish of 24 hpf(hour post fertilization, hpf) healthy TG fluorescence vascular system as experiment model, then process with kayaku oxaliplatin, 5-fluorouracil and Chinese medicine compound kushen separately, in combination and together for 24 hours to analyze indicator of intersegmental vessels and adopt SPSS to process experiment data. Utilize 48 hpf zebrafish embryos which have developed normally as model and add astemizole with density of 4 μmol/L, then add resveratrol and danhong injection with different density. After all of these procedures, observe change of embryonic heart shape, heart rate and sinus- arterial Ball(SV-BA).Results:The combination of kayaku oxaliplatin and 5-fluorouracil by 1 to 3 with Chinese medicine compound kushen injection have the best effects on inhibiting zebrafish segmental vessels; the compound Chinese medicine compound kushen injection is positively correlated with inhibition effects. Astemizole with density of 4 μmol/L will cause cardiac damage, including congestive heart failure, heart rate decrease and the distance between SV and BA is widened. Resveratrol with density of 60 μg/m L and danhong injection with density of 20 μL and 30 μL can restore cardiac damage caused by astemizole obviously.Conclusion:Kayaku oxaliplatin, 5-fluorouracil and Chinese medicine compound kushen injection alone, in combination and together all can inhibit intersegmental vessels of zebrafish. Chemical medicine and Chinese medicine together can strength effects. Resveratrol and danhong injection have restoration effects on cardiac injury caused by astemizole and can provide experimental basis for treatment on cardiovascular diseases.
