Objective:The purpose of the study was to detect the expression of protein Twist and the type 16 of Human papilloma virus(HPV 16)protein E6 in cervical cancer,cervical intraepithelial neoplasia and normal cervix,to evaluate their roles and clinical significance of Twist and HPV16 E6 in the carcinogenesis and development of cervical carcinoma,as well as the relativity of the two factors.Methods:Collecting pathological tissue 60 cases which had been removal of patients cervical tissue by surgical operation and been manufactured paraffin specimens.They are classified into three groups and consist of the normal cervical tissue 10 cases,cervical intraepithelial neoplasia(CIN)25 cases,cervical cancer tissue 40 cases.We will detect the expression of protein Twist and protein HPV16 E6 in cervical tissue by using immunohistochemical SP method and analysis of the relationship between the two characteristic parameters with clinical cases respectively and the correlation between them.Usingχ2 test comparing the differences of among counting data in three groups and Spearman method analysising the correlation between protein Twist and protein HPV16 E6 in cervical tissue.Results:In general,the expression of protein Twist lie in the cytoplasm or nucleus in the cervical tissue cell.Among in normal cervical tissue,CIN and cervical cancer tissues,the positive expression rate of Twist is 77.5%(31/40),36.0%(9/25)and 10.0%(1/10),expression showed aincreased trend gradually,and relative comparisions among three groups,the difference was statistically significant(χ2=22.644,P=0.000<0.05).The clinical parameters of patients with cervical cancer patients,such as age,FIGO staging,tissue differentiation degree and tumor size,the expression discrepancy of protein Twist had no statistical significance(P>0.05),but when patients with cervical cancer are divided into the groups of lymph node metastasis and without lymph node metastasis,Twist positive expression rate were 94.11%,65.22%,compared between two groups,the difference was statistically significant(χ2=4.682,P=0.030<0.05).2.In the CIN(16/25,64.0%)and cervical cancer group(35/40,87.5%),the positive expression rate of protein HPV16 E6 is significantly higher than normal cervical group(0/10,0.00%),and compared between three groups,the differences was statistically significant(χ2=28.424,P=0.000<0.05).In patients with cervical cancer,between different layers of pathological parameters,such as age,FIGO staging,tissue differentiation,tumor size,HPV16E6 protein expression differences have no statistical significance(P>0.05).(3)in cervical lesion,the positive expression between protein Twist and protein HPV16 E6 was positively correlated(rs=0.409,P=0.000<0.01)Conclusion:Both Twist and HPV16 E6 are involved in the occurrence and development of cervical cancer,and Twist promote the role in the process of tumor metastasis.Twist and HPV16 in the development of cervical cancer may play a synergistic action.
