| Objective:To research the expression of RNF43 in gastric carcinoma by the home-made tissue microarray; To explore its relationship with the clinical data of gastric cancer; To analyze the relationship between the expression of RNF43 and the prognosis of gastric cancer.Methods:A total of 144 gastric cancer patients in the First Affiliated Hospital of Shanxi Medical University from January 2010 to December 2013 were collected from patients of gastric cancer. Patients’ clinical datum is integrated, chemotherapy and radiotherapy were not performed before operation. The expression of RNF43 in 144 cases of gastric cancer tissues and adjacent tissues were detected by immunohistochemistry. The relationship between clinical data and postoperative survival rate were analyzed by statistical methods.Results:1. 5 blocks of TMA wax are built, and the wax stone are flat, no cracks, no bubbles,and the arrays were arranged in order. The organization is closely integrated with the wax block. The five tissue chips array is 10×6, the points are arranged in order, no obvious shift, no folding, no tissue loss and effective.2. RNF43 in gastric cancer tissues was mainly expressed in the cell nucleus. The positive expression rate in gastric cancer was 52.1%(75/144). The positive expression rate in the adjacent tissues of cancer was 34.0%(49/144).The group of two dates was statistically significant.3. The high expression of RNF43 in gastric cancer is related to the TNM stage, lymph node metastasis, differentiation degree, depth of invasion, and tumor size(P <0.05).There is no related to sex, age, microvascular invasion, nerve invasion, and tumor location(P>0.05).4. Relationship between gastric cancer RNF43 high expression and prognosis of gastric cancer using Kaplan Meier survival analysis was performed, the log rank test statistics have significance(P <0.05) in 144 patients with median survival of 47 months, RNF43 positive expression in gastric cancer survival period of 25 months,while negative expression survival > 50 months, thus, gastric cancer rnf43 expression than low expression of survival low.Conclusion:1. The expression of RNF43 in gastric carcinoma was significantly higher than that in the adjacent tissues, thus it can be seen that RNF43 was involved in the occurrence of gastric cancer.2. The high expression of RNF43 in gastric carcinoma was closely related to the TNM stage, lymph node metastasis, differentiation degree, depth of invasion, and tumor size.RNF43 may be an important marker for the metastasis and development of gastric cancer.3. The survival time that the high expression of RNF43 in gastric cancer was lower than low expression, and it is related to the prognosis of gastric cancer. Suggesting that RNF43 may be a marker of prognosis of gastric cancer.4. The self-made tissue microarrays can be according to the need of task, it can reduce the system error, saves time and improve test efficiency; The process of tissue microarrayis simple, easy to operate, and it is a highly efficient and feasible technology. |
