Objective:To observe the effect of Intermedin on endothelial-to-mesenchymal transition of renal fibrosis in UUO rat model.Methods:Seventy-two male Wistar rats were randomly divided into two groups:the sha m-operation group(n=24)underwent the left ureteral dissection,the other 48 rats were made as unilateral ureteral obstruction models and sub-divided into the model group(uuo),the IMD group(each group n=24). At the7,14,21,28 day after the operation,6 randomly-selected rats form each of the three groups respectively were taken out.Observe the glomerular pathology change.The TGF-β2,α-SMA,BMP-7,VE-ca dherin,VEGF and VEGFR2 on the expression of mRNA were detected by real time-PCR and immunohistochemistry was used to detect the expression of the protein(Including CD31 and JG-12).Results:Compared with the sham-operated group, the mRNA expression and protein content of TGF-β2,α-SMA increased(P<0.05),the levels of CD31,JG-12,BMP-7,VEcadherin,VEGF and VEGFR2 decreased(P<0.05);compared with the UUO group,the mRNA expression and protein content of TGF-β2, α-SMA were down-regula-ted(P<0.05), the levels of BMP-7,VE-cadherin,VEGF and VEGFR2 were up-regulated(P<0.05).Conclusion:IMD can ameliorate the glomerular fibrosis and protect renal function by restrain the effect of endothelial-to-mesenchymal transition of glomerular vascular endothelial cell In the process of renal fibrosis.
