Background And Objective: Chemokine receptor 3(CXCR3) is an ELR-negative chemokine receptor, which belongs to the G protein coupled receptor superfamily members, its expression has a very close relationship with tumor initiation, development and tumor metastasis. The expression of CXCR3 is significantly increased in breast cancer, renal cancer, colon cancer and ovarian cancer, and CXCR3 correlate with Tumor differentiation degree and grade. The research of the expression of CXCR3 in bladder cancer has not been found.The aim of this experiment is to investigate the role of CXCR3 in bladder cancer and its correlation with bladder cancer development, so as to provide new reference for bladder cancer clinical diagnosis and treatment.Methods: We collect 77 patients tissue of bladder cancer and some of their adjacent tissues(32 cases),and detect the CXCR3 expression differences between paracancer tissues and bladder cancer tissues and correlation between CXCR3 staining degree in cancer tissues with age,grade, stage by semi-quantitative immunohistochemical methods. 27 bladder cancer tissues and their corresponding paracancer tissues are kept in the RNA keeper, detecting the expression differences of CXCR3 and CXCR4, INF-γ mRNA between the bladder cancer tissues and the corresponding adjacent control tissues by RT-PCR.Result: The immunohistochemistry results showed that the degree of CXCR3 staining in bladder cancer tissues of patients was significantly higher than Paracancer(P <0.01), in cancerous tissue stained, the degree of staining of carcinoma was related to bladder cancer grade and stage.In the relatively high grade and stage tumor tissue, the degree of the immunohistochemistry stain was relatively high(P < 0.01). RT-PCR experiments results of 27 cases of bladder cancer tissues and the corresponding Paracancer tissue showed that CXCR3 and CXCR4 mRNA expression in bladder cancer tissues was significantly higher than that in the tumor adjacent tissues(P < 0.01),But there was no significant difference in the expression of INF-γ mRNA in bladder cancer tissues(P>0.05). Correlation analysis showed that there was a positive correlation between any two mRNA expression of CXCR3, INF-γ,CXCR4(all r value was positive, and all P value was less than 0.01).Conclusion: The experimental results indicate that there are some relevance between CXCR3 and bladder cancer development, and the expression of CXCR3 also plays an important role in the growth and metastasis of bladder cancer. CXCR3 may play a synergistic effect with CXCR4 to promote the initiation and development of tumor.
