Objective:To establish a mouse model of iron overload by intraperitoneal injection of iron sucrose and preliminarily investigate the impact of iron overload on the telomere length of bone marrow cell subsets through the measurement of the telomere length of the iron overload mouse model.Methods:1.A total of 21 C57BL/6 8 weeks old male mice were divided into control group and experimental group. The experimental group was received with iron sucrose injections at a dose of 200mg/kg bw every other day for 4 weeks(14 times),the control group received equal normal saline. During the experiment, one control group mouse and two experimental group mice were sacrificed every 2 to 3 weeks.2.Colorimetric method was used to measure the tissue iron concentration.3.Flow cytometry-Fluorescence in situ hybridization(Flow-FISH) was used to measure the telomere length of bone marrow cell subsets.Result:1.Compared with control group, the liver iron concentration of iron overload group was increased obviously, the heart iron concentration was increased as well.2.The relative telomere length(RTL) is diverse in different batches throughout the experiment, however, the tendency of the relative telomere length change can be observed through the comparison between the control group and the experimental group. The RTL of bone marrow CD4 positive cells seem shorten compared with control group after 12 weeks. The RTL of bone marrow CD8 a positive cells seem shorten at the forth and seventh week, but the shortness didn’t observed later. The RTL of bone marrow CD19 positive cells seem shorten at the forth and fourteenthweek. The RTL of bone marrow CD11 b positive cells seem shorten compared with control group after 14 weeks. The Sca-1 positive cells didn’t shorten during the experiment(14 weeks).Conclusion:1.The iron-overload mouse model was successfully established by intraperitoneal administration of iron sucrose with the dose of 2.8g/kg bw.2.Iron overload may shorten the telomere length of the bone marrow cells, and the shortness seem to happens earlier in CD4,CD8 a,CD19,CD11 b positive cell subsets.
