Objective: To study the antidiabetic effect of Taraxacum mongolicum components on alloxan-induced diabetic mouse model, to confirm the effective part, and study its effective mechanism of antidiabetic through the inhibition experiments of protein tyrosine phosphatase 1B and α- glucosidase, provide the basis information for clinic medicine; To further investigate the antidiabetic effect of Taraxacum mongolicum total polysaccharides combined with tiliroside on alloxan-induced diabetic mouse model, to provide the experimental basis for studying the new Chinese drug of antidiabetic. Methods: 1. Use water to extract the dry Taraxacum mongolicum and then filtrate it to get the total extraction, after then through the method of precipitation, ethanol extraction get the total Taraxacum mongolicum extraction and lipophilic component, hydrophilic component and polysaccharide component. 2. To establish the diabetic mouse model through inject of 65mg/kg alloxan from tail, the diabetic mice were divided into six groups randomly, and then given the double distilled water 、 metformin and Taraxacum mongolicum extraction respectively everyday, after 15 days gavage, use the body weight, fasting blood glucose(FBG), total cholesterol(TC), triglyceride(TG) as the detection index, to study the effects of Taraxacum mongolicum and its three components on suger and lipid metabolism in diabetic mice, select the effective part of Taraxacum mongolicum on hypoglycemic. 3. To study the anti-diabetic mechanism of Taraxacum mongolicum and its three components by inhibiting the activity of protein tyrosine phosphatase 1B and α-glucosidase. 4. To establish the diabetic mouse model through inject of 65mg/kg alloxan from tail, the diabetic mice were divided into five groups randomly, and then given the double distilled water, metformin, Taraxacum mongolicum polysaccharide solution, tiliroside, and the mixture of Taraxacum mongolicum polysaccharide and respectively every day, after 15 days gavage, use the body weight, FBG, FFA, total cholesterol(TC) and TG as the detection index, to study the combined effects of Taraxacum mongolicum total polysaccharide and tiliroside on antidiabetic in diabetic mouse. Results: 1. Each component effect on the alloxan-induced diabetic mouse, the results of antidiabetic experiment shown that each extraction components of Taraxacum mongolicum can improve the spirit gradually, increase the activity and the weight of diabetic mice, improve the symptom of “three higher one lower”, reduce FBG significantly and lower serum total cholesterol and triglyceride levels, relieve abnormal lipid metabolism. The Taraxacum mongolicum total polysaccharide had the best function, significantly different compare with control group(P<0.05), the antidiabetic function is equal to metformin group. This is illustrate that the Taraxacum mongolicum total polysaccharide is the effective part of Taraxacum mongolicum on antidiabetic. 2. The results of anti-diabetic activity screening experiments shown that the Taraxacum mongolicumtotal extraction and the Taraxacum mongolicum total polysaccharide have an inhibitory effect on protein tyrosine phosphatase 1B and α-glucosidase, and the total polysaccharide played the strongest function. 3. The Taraxacum mongolicum total polysaccharide group, tiliroside with Taraxacum mongolicum total polysaccharide group all can improve the spirit gradually, increase the activity and the weight of diabetic mice, improve the symptom of “ three higher one lower”, reduce FBG significantly and improve OGTT, reduce TC and TG, alleviate the abnormal lipid metabolism of diabetic mice. The effect of tiliroside with Taraxacum mongolicum total polysaccharide group is better than Taraxacum mongolicum polysaccharide significantly(P<0.05). Conclusions: Taraxacum mongolicum total polysaccharide can reduce blood glucose significantly, improve lipid metabolic disorder in alloxan-induced diabetic mice model, it is the effective part of the effective part of Taraxacum mongolicum on antidiabetic. Inhibited protein tyrosine phosphatase 1B and α-glucosidase maybe one of the mechanism. Combined Taraxacum mongolicum total polysaccharide and tiliroside significantly reduced blood glucose level, regulated lipid metabolism disorder on alloxan-induced diabetic mice, suggested that the combination of Taraxacum mongolicum total polysaccharide and tiliroside has a synergistic effect。...
