Objective:with detection of expression level miR-193a-5p, mir-486-5p, miR-4778-5p, miR-4271, miR-551b-3p, miR-4646-5p in Xuanwei lung adenocarcinoma, analysis of its clinical significance, biological information analysis to lays the theoretical foundation for the follow-up mechanism study of miRNAs in Xuanwei lung adenocarcinoma.Methods:using snRNAU44 and snRNAU48 as the internal control for real-time fluorescence quantitative PCR in detecting expression level of six miRNAs in 35 paired cancer and corresponding normal Xuanwei lung adenocarcinoma tissues. Correlation ananlysis of log2 transformed folde change of six miRNAs and clinical data then were carryed out. Genomic location analysis of miRNAs precursor with Ucsc genome database and 10KB upstream possible promoter region were selected, predicted promoter with neural network promoter prediction database, predicted transcription start site with patch database predicted possible transcription factor with promoter 2.0 and PROSCAN, predicted methylation sites with EMBOSS database were conducted.The targetscan, DIANA-TarBase, Miranda database were used to predicte potential target genes,GO analysis of thepotential target genes were used to suggeste role of the six miRNAs in Xuanwei lung adenocarcinoma may plays.Results:regardless snRNAU48 or snRNAU44 as reference miR-193a-5p, mir-486-5p, miR-4778-5p, miR-4271, miR-551b-3p and miR-4646-5p were down regulated (p less than 0.05) in Xuanwei lung cancer tissue compared with corresponding normal tissues. Logarithmic transformated folde change expression change ratio less than 1 had no relation with the age of patients is greater than 60 years of age, tumor size is less than 1cm, tumor infiltration of growth, tumor occurrence of regional lymph nodes metastasis. Bioinformatics analysis showed the promoter region methylation, SP1 transcription factors may be involved in theRegulation of six micrornas. miR-193a-5p, mir-486-5p, miR-4778-5p, miR-4271, miR-551b-3p, miR-4646-5p target genes is involved in DNA methylation, regulation of cell DNA biosynthesis, regulation of cell membrane stability, participation in regulation of tumor microenvironment, participation in cell signal transduction, involved in cell migration and apoptosis etc.Conclusion:MiR-193a-5p, mir-486-5p, miR-4778-5p, miR-4271, miR-551 b-3p, miR-4646-5p were down-regulationed in Xuanwei lung adenocarcinoma and the six miRNAs may via target gene taking party in cell signal transduction, regulation of tumor microenvironment, involved in the cell transfer, regulation of cell DNA biosynthesis, involved in cell apoptosis, regulation of cell membrane stability, involved in DNA methylation. This experiment laid the theoretical basis for further mechanism study of these six miRNAs in Xuanwei lung adenocarcinoma and explored new ideas.
