Objective:Changtai is an empirical formula for the treatment of colorectal cancer in Affiliated Hospital of Integrated Traditional Chinese and Western Medicine, Nanjing University of Chinese Medicine. In this paper, we analyzed the quality control of changtai by HPLC. A heterotopic transplantation tumor model was established to investigate the inhibition effect of Changtai on colorectal cancer and explore its mechanism.Method:1) Analysisthe quality control of changtai by HPLC.2) A heterotopic transplantation tumor model was established to investigate the inhibition effect of changtai on colorectal cancer, the tumor volume (mm3) was estimated using the equation length×(width)2×0.5. The cell apoptosislevels of tumor tissues on tumor-bearing mice were detected by one-step Tunel stain.3) Western blotting was used to detect protein levels of Cleaved caspase-9, Cleaved caspase-3,PARP, Cleaved PARP, Bcl-XL, Bax,LC3I, and LC3Ⅱ from tumor tissues.Results:1) Acrodingto the external standard method, changtai consisted of 0.273mg/g Ginsenosides Rgi,0.457mg/g Matrine,0.492mg/g Ginsenoside Rbi,0.791mg/g Astragaloside,0.008mg/g Ferulic acid and 0.166 mg/g Calycosinglycosidesof crude drug.2) Changtai could significantly inhibit transplanted tumor growth in nude mice. The growth inhibition rate was significantly increased with increased concentration of changtai, as well as the cell apoptosis levels detected by TUNEL.3) Changtai can activate Caspase-9, Caspase-3 and PARP, increase the expression of pro-apoptogenic factor Bax, inhibit the expression of anti-apoptogenic factor Bcl-XL.Changtai can increase the expression of autopragy protein LC3.Conclusions:1) HPLC analysis can be used for the quality control of changtai.Changtai consisted of 0.273mg/g Ginsenosides Rg1,0.457mg/g Matrine,0.492mg/g Ginsenoside Rb1,0.791 mg/g Astragaloside,0.008mg/g Ferulic acid and0.166 mg/g Calycosin glycosidesof crude drug;2) Changtai could significantly inhibit transplanted tumor growth in nude mice and promote tumor cells apoptosis;3) Changtai could induce autophagy and cell apoptosis in human HT-29 transplanted tumor tissue in nude mice.
