Objective1. To observe the changes of skin lesions and pathological in mice2. To investigate the effects of Xiaoyin Mixture on Th17 cytokines in psoriasis-like mice model.3. To explore the possible mechanisms of Xiaoyin Mixture to treat psoriasis.Methods45 BALB/c mice were randomly divided into 5 groups, each group contained eight mice.The groups were:the model group, blank control group, Xiaoyin Mixture of high and low dose group, MTX group.The rest 5 mice were for preliminary experiments.Go on with the experiment after the evaluation model method was successful.Except the blank control group with vaseline, the rest 4 groups with 5% imiquimod cream 42mg to mouse back skin everyday.Each group of mice were treated with the drug orally:the blank control group, model group received normal saline. Xiaoyin Mixture of high and low dose groups with stomach perfusion once a day, for 12 days.Observe the mouse dorsal skin for PASI score; collect blood through the eyeball and than detect cytokines in serum;take back pathological tissue sections for relatively organizational research.Throughout the whole experiment, watch the mice closely.Results1. Mice treated by topical imiquimod(IMQ) showed typical psoriasis-like skin lesions such as erythema, scales, and thickening. There was a parabolic change of PASI scores. Compared with the mice of model group, Mice of low-dose, high-dose Xiaoyin Mixture groups and MTX group showed slight changes in both skin lesions and PASI scores.2. Compared with model group, skin lesions level of IL-17、IL-22、IL-6、TNF-α were significantly lower in low-dose, high-dose Xiaoyin Mixture groups and MTX group.3. HE staining showed epidermal parakeratosis, hyperkeratosis, and acanthosis in mice of model group. Compared with model group, epidermal thickness was significantly lower in low-dose, high-dose Xiaoyin Mixture groups and MTX group.ConclusionsXiaoyin Mixture could significantly impede the arising of the psoriasiform skin lesions and pathological changes induced by imiquimod, and significantly lower he level of Th17 cytokines and chemokines.
