Objective:To detect the expression of Angptl 3 protein and Angptl 4 protein in invasive breast cancer and breast fibroadenoma,and analyze their relationship with clinical pathology characteristic in breast cancer, assess their impact on the prognosis of breast cancer, explore the significance of them in breast cancer in order to provide valuable biomarkers for determining the treatment and prognosis.Method:The expression of Angptl 3 and Angptl 4 were respectively measured in 70 breast cancer tissue samples and 30 breast fibroadenoma samples by immunohistochemical SP method. SPSS 17.0 statistical software was used to analyze the relationship between the two proteins and the age, tumor size, axillary lymph node metastasis and histological grade, pathological staging (pTNM), pathological type, estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER-2) and prognosis of the disease, and to analyze the correlation between the expression of Angptl 3 and Angptl 4 in invasive breast cancer.Result:1、Immunohistochemical results showed that the positive rate of Angptl 3 in breast cancer and breast fibroadenoma were 75.7%(53/70)and 46.7%(14/30)(X2=8.014,P=0.005).The Angptl 4 positively was observed in 68.6%(48/70) and 36.7%(11/30)in the two groups of breast cancer and breast fibroadenoma (X2=8.837,P=0.003).The difference of Angptl 3 and Angptl 4 expression between breast cancer and breast fibroadenoma was significant.2、The expression of Angptl 3 was not correlated with age, histological grade, histological grade, PR, and Her-2 (P>0.05), but has significant correlation with tumor size, axillary lymph node metastasis, TNM stage, and ER (P<0.05). The expression of Angptl 4 was not correlated with age, histological grade, histological grade, ER, PR, and Her-2 (P>0.05), but has significant correlation with tumor size, axillary lymph node metastasis, TNM stage (P<0.05)3、In 70 cases of invasive breast cancer,There were 41 cases of positive expression of Angptl3 and Angptl4 at the same time, there were 10 cases of negative expression, Angptl3 expression was positive while Angptl4 expression was negative in 12 cases, Angptl4 expression was positive while Angptl3 expression was negative in 7 cases. The expression of Angptl3 and Angptl4 was positively correlated with Spearman rank correlation analysis (P=0.005, r=0.334).4. Meier Kaplan survival analysis showed that patients with negative expression of Angptl3 and Angptl4 protein were better than those of protein expression in the overall survival period, (P<0.05). Cox multivariate survival analysis showed that age, histological grade, tumor size, and ER were independent risk factors affecting prognosis (P<0.05), while the Angptl3 and Angptl4 protein expression did not enter the Cox regression model (P>0.05).Conclusion:1. Compared with breast fibroadenoma, Angptl 3 and 4 in breast cancer was higher, and this two kinds of protein expression was positively related and jointly play a cancer promoting effects.2. High expression of Angptl protein were related to the bigger tumor size, advanced TNM stage, lymph node metastasis in breast cancer patients. But only Angptl 3 was higher in ER positive patients.3. The survival period of patients with Angptl positive expression was shortened, So this two proteins are the factors that affect the prognosis of invasive breast cancer, but multivatiate COX regression analysis showed that the expression of protein was not independent affecting factor to breast cancer patients.
