It is a wildly held view that osteoarthritis (OA) is an degenerative disease which generates degradation of cartilage as its central pathological process.Objective:In this research, Licochalcone A, was investigated about its anti-osteoarthritis effect in rat chondrocyte pretreated by interleukin-1β.Methods:qRT-PCR and western bloting were performed to scrutinize the expressions of Collagen type II (Collagen II), matrix metalloproteinase (MMP)-1, MMP-13, A Disintegrin And Metalloproteinase with Thrombospondin Motifs (ADAMTS)-4 and ADAMTS-5 in both gene and protein levels. Meanwhile, the activations of Wnt/β-catenin and NF-κB signal pathways were analyzed quantificationally by western bolting. The findings of our study suggested that Licochalcone A inhibits the degradation of Collagen Ⅱ with down-regulation of the MMP-1, MMP-3, ADAMTS-4 and ADAMTS-5, and the amount of P-catenin and the phosphorylation of p65, IKKα/β is suppressed simultaneously.Results:The findings of our study suggested that Licochalcone A inhibits the degradation of Collagen Ⅱ with down-regulation of the MMP-1, MMP-3, ADAMTS-4 and ADAMTS-5, and the amount of β-catenin and the phosphorylation of p65, IKKα/β is suppressed simultaneously.Conclusions:We can infer that Licochalcone A inhibits MMPs and ADAMTSs via p65-dependent NF-κB pathway together with Wnt/β catenin signaling in rat chondrocytes. Thus Licochalcone A could be used as an anti-inflammatory reagent in the treatment of OA and should be further studied. |
