Hypertrophic scar and keloid in the clinical have occurred sometimes. Studies have indicated that the TGF-β pathway and the Toll like receptors have important effects on the proliferation of fibroblasts and the secretion and deposition of extracellular matrix in the process of scar formation and progression. At the same time, in the process of scar formation and development, fibroblasts are suffered the state of hypoxia stress. Study shows that hypoxia inducible factor, HIF in tumor and organ fibrosis, there is interaction and influence between HIF and TGF-β pathway and TLRs. This study through the construction of human fibroblasts in vitro HFF-1 hypoxia model and validated this model by Western Blot, immunofluorescence and biochemical methods to detect changes of fibroblast in vitro expression of TGF-β 1, TGF-β RⅡ pathway and TRLs related proteins. The results showed that after 1%O2,5%CO2 cultured 24h, HFF-1 cell into hypoxia stress. And the expressions of Smad2/3, Smad4, Smad7, TLR4, TLR7, ERK1/2, CTGF, ERK1/2 in cell are increased. And the phosphorylation level of Smad2/3 and ERK1/2 increased in hypoxia group. The TGF-PRII shows more at cytomembrane after the hypoxia cultured. Collagen intracellular synthesis and deposition increased, and extracellular free collagen decreased. The results showed that the hypoxia stress model for HFF-1 was successfully constructed in this experiment. The hypoxia stress have shown the effects on the TGF-β pathway, TLR4, TLR7 and the related proteins in the downstream.
