The mortality of gastric cancer ranks third in the world. The lymph node metastasis molecular mechanism of the disease is a hot spot. In previous research, people found that phospholipase A2 (PLA2) was closely related to the proliferation of gastric cancer. cPLA2 is the main effective subtype of PLA2. Phospholipase A2-IVA (cPLA2G4A) is the most abundant subtype of cytosolic phospholipase A2 (cPLA2). The role of cPLA2G4A in the migration and progression of gastric cancer is not clear.Objective:TO observe the effect of AACOCF3 on the proliferation and mobile ability of gastric cancer cell lines BGC823, SGC7901, MKN45. To elucidation the expression and the role of phospholipase A2-IVA (cPLA2G4A) in gastric cancer,and the association of phospholipase A2-IVA (cPLA2G4A) with cyclooxygenase 2(COX-2).Methods:Culture cell line regularly, MTT colorimetric method was used to detect the proliferation of the three cell lines. Mobile ability was detected by the trans-well at different time points (24h,48h,72h). We collected pathological specimen of gastric carcinoma, serum sample from healthy subjects and gastric cancer patients.Serum concentration of phospholipase A2-IVA (cPLA2G4A) in healthy subjects and gastric cancer patients were detected by ELISA.Results:The proliferation of gastric cancer cell were detected in three time points and different concentrations. With the extension of time and concertration, the proliferation of BGC823, SGC7901, MKN45 were decreased gradually. In Trans-well experiments we found that the migration and invasion ability of these three kinds of gastric cancer cell were inhibited after using AACOCF3. The concentration of phospholipase A2-IVA (cPLA2G4A) in gastric cancer patients’ serum were higher than that of healthy samples (P<0.05). Phospholipase A2-IVA (cPLA2G4A) was significantly associated with tumor grade(P<0.05), N classificatin(P<0.05).Conclusion:1. AACOCF3 can inhibit the proliferation and motile ability of BGC823, SGC7901, MKN45. cPLA2G4A combined with COX-2 might serve as a target for gastric cancer therapy.2. cPLA2G4A in gastric cancer was closely related to the tumor grade and lymph nodes metastasis.
