Effects Of Wenyang-Huoxue-Jiedu-formula On The MRNA Expression Of VEGFs/VEGFRs Pathways In HUVECs And Clinical Cases

ObjectiveAcute coronary syndromes (ACS) are the leading cause of the urgency. Modern theories of atherosclerosis (AS) mainly focus on the angiogenesis in the thin-caP fibroatheroma Plaques (TCFA). Recent studies have shown vascular endothelial growth factors and its receptors (VEGFs/VGFRs) enhance the formation and progression of TCFA. The conception of inflammation is same to Duxie in TCM, so we consider Wenyang-Huoxue-Jiedu-formula (WHJF) is effecti-ve in ACS therapy. Our previous research found that WHJF arrested the proliferation, migration, lumen formation and the VEGFs /VGFRs expression of human umbilical vein endothelial cells (HUVECs). In the present study, we are trying to study the effects of WHJF on the mRNA expression of VEGFs/VGFRs and evaluate the effectiveness of the therapy on clinical cases.MethodsHUVECs were bought from a stable line and routinely cultured in DMEM supplemented with 20% fetal bovine serum (FBS) at 37℃ and 5% CO2. SD mice were fed on saline, which served as control group, and low-, middle-, high-dosage of the formula. WHJF serum was obtained on the fourth day at 2 hour after first administration. Cells were treated with a concentration of 10% of the medication-contained serum for 12 hours. The mRNA expression of signaling molecules in VEGFs/ VEGFRs pathways were assayed by quantitative real-time polymerase chain (qRT-PCR).The clinical cases of ACS intended to study were randomly divided into the control group and the experiment group. The patients in the control group were treated with routine medications, while the experiment group with routine medication combined with WHJF. The red blood cell distribution width (RDW), hospital mortality, questionnaire and effectiveness of Chinese medical syndrome were used to evaluate the effectiveness of these two therapies.ResultsThe serum of WHJF has shown no effects on the mRNA expression of VEGFs/ VEGFRs, though it has the ability of down-regulating the expression of VEGFs/ VEGFRs. The results of the clinical case have revealed that the experiment group has better effect than the control group. The RDW of experiment group was lower than the control group’s ones. The hospital mortality between the groups has no differience.ConclusionWHJF shows no effects on the mRNA expression of VEGFs/ VEGFRs, though it prevents angiogenesis via down-regulating the mediate molecules. WHJF maketly improves the effectiveness of the patients with ACS.
