Objective:To discuss the cultured gallbladder carcinoma cells in vitro, the correlation between Heparanase gene to over-express and Syndecan-1 expression with the invasive ability of gallbladder carcinoma cells.Method:1. In order to observe the growth features in vitro and draw the cell growth curve,Heparanase gene to over-express of GBD-SD and normal expression of GBC-SD to culture,passage and freeze in vitro.2. Western-blot methods are applied to detect the protein expression of Syndecan-1 in GBD-SD and GBC-SD.3. RT-PCR technologies are applied to detect the Syndecan-1 gene expression quantities in GBD-SD and GBC-SD.4. Transwell cell methods are applied to detecting the invasive ability of GBD-SD and GBC-SD separately.Result:1. Not only recovered GBD-SD and GBC-SD, made it’s subculture and cryostorage successfully, but drawed the cell growth curve and realized the gallbladder carcinoma cells growth character in vitro as well.2.The expression of Syndecan-1 protein in GBD-SD was decreased sharply compared with Syndecan-1 in GBC-SD, and the difference was statistically significant(P<0.05).3. The m RNA of Syndecan-1 in GBD-SD significantly reduced than that of GBC-SD, the difference was statistically significant(P<0.05).4. The number of cell invasion of GBD-SD was significantly more than that of GBC-SD,and the first one showed a stronger invasive ability. The difference was statistically significant(P<0.05).Conclusion:1. In gallbladder carcinoma cells, the over-expression of Heparanase gene and the expression of Syndecan-1 presents negative correlations, and the invasive ability of gallbladder carcinoma cells and the over- expresion shows positive correlation.2. In gallbladder carcinoma cells, the expression of Heparanase was increased, the expression of Syndecan-1 was decreased, and the invasive ability was enhanced, and the Heparanase/Syndecan-1 axis played an important role in the invasion and infiltration of gallbladder carcinoma.3. Heparanase/Syndecan-1 axis is studied for the diagnosis and treatment of gallbladder cancer provides new ideas and methods. At the same time, as the development to axis of Heparanase/Syndecan-1 target drugs to provide new theoretical and experimental basis, but axis of Heparanase/ Syndecan-1 of gallbladder cancer biological behavior affects the detailed molecular mechanisms, it’s up in the air so we need to study further more.
