| Objective:Formaldehyde (FA) is a kind of highly active air pollutant that widely used in factories and other sides. It can cause multiple diseases and organ toxicity. A large number of epidemiological data shows that FA may cause leukemia, particularly myeloid leukemia, so as the animal studies show. However, the specific pathogenesis is unclear. Leukemia is the malignant tumor that occurs at hematopoietic system, the proliferation and edifferentiation of hematopoietic stem cells regulated by a lot of factors such like the cytokines. Benzene (BZ) is a kind of air pollution that can cause hematopoietic toxicity, always appears at the same time with the FA whether in the environment or factories. Thus, to study the number of blood cells and cytokines that regulation of hematopoietic system, to explore the hematotoxicity that FA expose with or without benzene in mice.Methods:30 males SPF BALB/mice, were randomly divied into 5 groups, six of each group. The specific way of grouping and infected animal experiments:(1) the blank control group (Ctrl); (2)the corn oil control group (Oil):with concentration of 5ml/kg bw/day corn oil to fill the stomach;(3) the benzene separate exposed group (BZ): with concentration of 150mg/kg bw/day of benzene (corn oil as solvent) to fill the stomach;(4) the formaldehyde separate exposed group (FA):to inhale the formaldehyde gas with concentration of 3.0 mg/m3;(5) the benzene and formaldehyde jointly exposed group:in the morning of the expose days, to fill the stomach of the benzene (corn oil as solvent) with concentration of 150mg/kg bw/day first, then inhale the formaldehyde gas with concentration of 3.0 mg/m3, the way, time and concentration of lavage is the same with the BZ group, the expose time, concentration and mode of inhaling gas formaldehyde is the same with the FA group. The expose time is five days a week,8 hours a day for two weeks. After the expose cycle, put the mice to death, then measure and observe a series of index of mice.Results:1.Each group of mice weight generally presents the rising trend, but the increase margin of formaldehyde with the exposed group is relatively slow, it indicated that formaldehyde expose can effect the growth and development of mice.2.Under the effect of formaldehyde canister to separate, the white blood cell, granulocytes, lymphocyte and monocyte giant cells have no obvious change. And benzene separately and the combination of formaldehyde and benzene, the number of these cells were significantly decrease (P<0.01), (relatively The Ctrl group and Oil group). However, the number of red blood cells are roughly unchanged, benzene group work alone relatively the Oil group decreased significantly (P<0.01).3. The formaldehyde separate action can make the cytokine M-CSF transcription level decreased (P<0.01), with benzene, benzene strengthen formaldehyde affect cytokines M-CSF.4. The formaldehyde separate action can make the cytokine G-CSF transcription level decreased (P<0.01), with benzene, benzene weaken formaldehyde affect cytokine G-CSF.5. The formaldehyde separate action can make the cytokine EPO transcription level decreased (P<0.01), with benzene, benzene weaken formaldehyde affect cytokine EPO.Conclusion:under a certain concentration, formaldehyde alone can certain blood toxicity in mice, the benzene to exit, can weaken the toxicity effect of formaldehyde. |
