Objective: This study aims to analyze the anti-inflammatory action of Clausena lansium(Lour.) Skeels extracts and their inhibition effect on LPS-induced inflammation via TLR4/My D88/TRAF6 signaling pathway.Methods: After treated with lipopolysaccharide(LPS) at different times, using ELISA and RT-PCR measures to detect the TNF-α. After Clausena lansium(Lour.) Skeels extracts treated RAW264.7 cell, using ELISA and RT-PCR measures to analyze TNF-α. Using MTT measure to analyze whether Clausena lansium(Lour.) Skeels extracts with different concentrations and LPS have affect on the viability of RAW 264.7 cells. According to the experiment,RAW264.7 cells were divided into five groups: control group, LPS+5 m M MyD88 inhibitor group, LPS group, LPS+ extracts group and LPS+ extracts+5mM MyD88 inhibitor group. RT-PCR and Western Blot were applied to measure TLR4, My D88, TRAF6 and TNF-α m RNA and protein. Results: The results indicate that when different concentrations of Clausena lansium(Lour.) Skeels extracts(25、50、100ug/mL) were combined with 1 ng/mL LPS, there were no obvious influence on the cell viability of RAW264.7 cells. RAW264.7 cells were exposed to different concentrations of LPS(0.1, 1, 10, 100 ng/mL), the results indicated that LPS induced TNF-α secretion, with the level of TNF-α reaching a peak value of 1ng/mL. Using the concentration of 1 ng/mL LPS has stimulatedRAW264.7 cells, with the increase of time(4, 8, 12, 24, 48h), TNF- a secretion increases;and Clausena lansium(Lour.) Skeels extracts inhibited the secretion of TNF-α in a dose-dependent manner. furthermore, My D88-dependent signaling pathway might be involved in the mechanism of Clausena lansium(Lour.) Skeels extracts to suppress LPS-induced inflammation, MyD88 inhibitor enhances the effect. Conclusions: The Clausena lansium(Lour.) Skeels extracts inhibit LPS-induced inflammation in RAW264.7 cells. TLR4/MyD88/TRAF6 signaling pathway might be involved in the mechanism of Clausena lansium(Lour.) Skeels extracts to suppress LPS-induced inflammation and My D88 inhibitor coupled with the Clausena lansium(Lour.)Skeels extracts synergistically enhanced the Antisecretory effect.
