Objective: To investigate the effect of COH by GnRHa on the Ezrin protein and mRNA expressions of mice endometrium during implantation window, then expolre the effect of COH on mice endometrium during implantation window, and further illustrate the relationship between controlled ovarian hyperstimulation restrained endometrial receptivity and the effect of COH on mice endometrium during implantation window. Methods: 60 SPF unfertilized female Kun Ming mice(7 to 8 weeks) were randomly allocated into 2 groups, control group and COH group. Mice in COH group treated with intraperitoneal injection of GnRHa 0.4 μg/g at 9:00 am for a consecutive 9 days in metestrus, with the mutual mix of HMG 0.01 IU/g at the ninth day, h CG 1 IU/g injected after 48 hours. And mice of control group were treated with the same volume of saline. Vaginal fluid smear was applied after drug used, periodic variation decreased even disappeared suggested pituitary desensitization, then recovery of periodic variation injected with HMG indicated successful modeling. After successful modeling, mice caged by the ratio of 2:1(male:female), thrombus were examined every noon, mice with thrombus marked first day of pregnancy. 0.3 ml 0.5% evans blue was injected via tail vein at the forth day of pregnancy and then mice were sacrificed and laparotomize to take uterus and ovaries for immunohistochemistry/ detection of mRNA and Western-blot. Results: 1. Cvaginal plug rate/pregnancy rate and implantation rate of embryos :Cvaginal plug rate / pregnancy rate was reduced in COH group; implantation numbers of embryos in COH was higher, but implantation rate of embryos was lower, normalized to control. 3. Result for immunohistochemistry: Protein expression level of Ezrin was greatly improved in COH group and mainly concentrated in progesterone-primed luminal epithelium and cytoplasm and cytomembrane of epithelia cells during mice endometrium implantation window, compared to control. 4. RT-PCR indicated mRNA expression level of Ezrin during mice endometrium implantation window was significantly higher, compared to control. 5. Western-blot suggested Ezrin expression was greatly improved compared to control. Conclusions: Ezrin was located in progesterone-primed luminal epithelium and cytoplasm and cytomembrane of epithelia cells of mature female mice; COH might restrain endometrial receptivity via improving expression level of Ezrin in ovaries.
