ObjectiveIn order to investigate the role of homemade apple enzyme in the therapy of diabetes, the type Ⅱ diabetic mice model induced by the streptozocin and high fat diet was constructed and the effects of homemade apple enzyme on reducing blood glucose, triglyceride and repairing pancreatic. To find a new, long-term, and safe prevention and treatment of diabetes and to provide a theory basis.Methods1. Totally 70 Kunming male mice with 8 weeks old and 42±2.93 g were randomly selected. Random 70 mice can be divided into normal control group(n=10),type Ⅱ diabetes mellitus model control group(n=12), low dose of apple enzyme group(n=12), middle dose of apple enzyme group(n=12), high dose of apple enzyme group(n=12), positive control group(metformin)(n=12).2. The normal control group was induced by five intraperitoneal injection of critic acid buffer solution after four weeks on normal diet. Experimental type Ⅱ diabetes was induced by five intraperitoneal injection of Streptozotocin(70mg/kg)after four weeks on high fat diet. The development of hyperglycemia in mice was formed by fasting blood glucose estimation after one week of STZ injection. The fasting blood glucose of any animal that were well maintained at a level higher than 11.1mmol/L were considered diabetic for studies.3. High dose of apple enzyme group, middle dose of enzyme group, low dose of enzyme group were intragastric administration with homemade apple enzyme once every day for 4 weeks. Thus, normal control group and type Ⅱ diabetes mellitus model control group were intragastric with sterilized distilled water. We detect the level of blood glucose of mice in each group and the weight of mice every week.4. At the end of the experiment, the mice were fasted for 4 hours then received oral glucose tolerance test(OGTT), we need to observe the changes of the area under the curve of blood glucose. Then enucleation of eyeball blood samples were performed for measurement of triglyceride(TG), total cholesterol(TC). At last, the mice were killed by cervical dislocation, pancreatic pathology was studied by morphological methods.5. Mean and standard deviation( x ±s), and the analysis of variance were used to compare between groups.Results1. The weight of the normal control group was stable. The weight of the others were decreased, especially the early stage of the model groups were relatively fluctuant,and the latter were in a relatively stable level. At the same time, different degree of depression and increase of urine volume were appeared.2. Different intervention were accepted after 2 weeks in each group. Compared with the model control group, the behavior of apple enzyme treated groups and positive control group mice were gradually active, having a good mental condition and reduction in urine volume. The fasting glucose level of metformin treated group decreased. Compared with the model control group, the fasting glucose level, levels of TC and TG in this group were lowered remarkably, with significant difference(p<0.05).3. Influence on glucose tolerance in mice, each group compared with model control group, 2h after lavage, we can find that apple enzyme reduce the rise in blood glouse levels in mice, the difference is statistically significant(p< 0.05).4. Compared with those of the normal control group, the effection of apple enzyme with high, middle and low dose treated group on pancreas tissue were similar,suggesting that enzyme has an effect on repairing pancreatic.ConclusionsApple enzyme can effectively decrease blood glucose, triglyceride, total cholesterol and repair pancreas of type Ⅱ diabetic mice.
