Objective1.To investigate the molecular epidemiological characteristics of HAd V in Changsha area during 2012-2014, and provide theoretical basis for the prevention and control of HAd V.2. The homology and difference of structural protein genes of human adenovirus 14, 55 and 57 in Changsha area were compared and analyzed,which provided experimental evidence for the evolution of HAd V.Methods Human Adenovirus positive samples were collected from patients with acute lower respiratory tract infection in hospitalized children from April2012 to March 2014 in Changsha area, which were nasop Haryngeal aspiration and bronchoalveolar lavage fluid samples. The samples were identified as human adenovirus type 14, 55 and 57 by PCR technique.Cell culture technique was used to isolate and culture adenovirus, and the virus’ nucleic acid was extracted. Fiber, hexon, penton structural protein genes were amplified by PCR method, and 14, 57 type whole genome were sequenced. The gene sequences were compared and analyzed by Meg4, Aligmen and DNAman, and the phylogenetic tree was constructed.Result1. In this study, 221 samples were successfully divided into three subgroups: B, C, and E.And 112 samples for HAd V-7(50.68%),26 forHAd V-1(11.76%),25 for HAd V-2(11.31%), 22 for HAd V-3(9.95%), 11 for HAd V-4(4.98%),7 for HAd V-5(3.17%),9 for HAd V-14(4.07%),3 for HAd V-6(1.36%),3 for HAd V-57(1.36%),2 for HAd V-55(0.90%),1 for HAd V-21(0.45%).2.There are base mutations in the coding region of Human Adenovirus type57: the presence of individual amino acid substitutions and variants in fiber and penton.3.The whole genome of human adenovirus type 14, which has high homology with the gene samples at home and abroad in recent years, and no base mutation in the structural protein gene region.4. The similarity of penton genes of HAd V- 14 and HAd V- 55 in Changsha, Chenzhou and Lanzhou regions was 100%, and the similarity of multiple samples in Genbank was 100%. And there was no difference in penton gene between multiple samples in Genbank and in Changsha area.5. The similarity of fiber gene of HAd V-55 in Changsha and Chenzhou was 100%, but the similarity was 99% in Lanzhou.And there was no difference in fiber gene between Samples from Changsha regions and Genbank in different regions and different times. The similarity of fiber gene of HAd V-14 of each sample in Changsha area was 100%, and the similarity of most of the domestic samples in Lanzhou area and most ofthe Genbank was also 100%,and the similarity of the individual samples in Lanzhou and Chenzhou area is 99%6. The similarity of hexon gene of HAd V-55 with Chenzhou, Changsha area Lanzhou area is 100%, and the majority of the samples in Genbank were also 100%.The similarity of hexon gene of HAd V-14 of each sample in Changsha area was 100%, and the similarity of most of the domestic samples in Chenzhou area, Lanzhou area and most of the Genbank was also 100%.7. The difference of hexon gene between Human Adenovirus type14 and Human Adenovirus type55 was 8%, which was the main difference between these two types of adenovirus, and the difference between fiber and penton gene was only 0-0.5%;Conclusions During the study period, changes in the main epidemiological type HAd V in Changsha area, the main epidemic strains was type 7, type 1, type2.The genome of HAd V-14 is relatively stable; HAd V-57 genome has the characteristics of time and space evolution.The prevalence of HAd V-55 and HAd V-14 structural protein gene was relatively stable in Changsha area.
