Dayuanyin( Composed of Areca catechu, Magnolia officinalis Rehd. et Wils., Anemarrhena asphodeloides Bun, Glycyrrhiza uralensis Fisch, Amomum tsaoko, Paeonia lactiflora Pall., Scutellaria baicalensis Georgi.) is derived from “The theory of the plague”, which is written by Youke Wu. It is used to cure virus infectious fever, dampness-heat, some unexplained fever, night sweats, viral encephalitis and so on.This paper study on the therapeutic effect of Dayuanyin on lipopolysaccharide-induced(LPS-induced) acute lung injury(ALI) in mice and to determined the active fraction of Dayuanyin. The therapeutic effect mechanisms of Dayuanyin was explored in terms of inflammatory cytokines, complement levels.Anti-complementary activity test in vitro indicated that the water extract of Dayuanyin(DYY-H2O), alcohol precipitation of the supernatant(DYY-CSL) and ODS eluted 4, 5 part(DYY-4, DYY-5) showed the inhibitions on the complement system towards the classical pathway.DYY-H2 O could decrease LPS-induced pulmonary edema and pulmonary tissue injury; DYY-CSL could also decrease acute lung injury induced pulmonary edema and pulmonary tissue injury; DYY-4 and DYY-5 could decrease the LPS-induced acute lung injury.DYY-H2 O, DYY-CSL, DYY-4, DYY-5 could reduced the mouse acute lung injury by induced lipopolysaccharide pulmonary edema and the lung tissue damage through decrease lung wet-to-dry weight(W/D) ratio, protein concentration in bronchoalveolar lavage fluid(BALF), down-regulation the level of tumor necrosis factor-α(TNF-α), interleukin-6(IL-6) and interleukin-1β(IL-1β) and up-regulated the level of interleukin-10(IL-10) in ALI model, decreased the level of C3 and C5 a in serum, attenuated lung histopathological changes and significantly reduced complement 3c deposition. Additionally, DYY-4 and DYY-5 also decreased the level of interleukin-4(IL-4) and interleukin-13(IL-13) in BALF, the C3 c level in serum.By means of silica gel column, Sephadex LH-20 gel column, recrysta-llization, P-HPLC and so on, 20 compounds were isolated from DYY-4 and DYY-5, their structures were identified by their physico-chemicalcharateristics and detailed spectroscopic analysis. 12 compounds were identified as wogonin, glycycoumarin, wogonin 7-O-β-D-glucuronide methyl este, liquiritigenin, skullcapflavone Ⅱ, isoliquiritigenin, glyasperin F 、licoisoflavone B, daucosterol, glycyrol, Magnolol, honokiol. As chemical analysis showed, DYY-4 and DYY-5 mainly from Glycyrrhiza uralensis Fisch, Magnolia officinalis Rehd. et Wils, Scutellaria baicalensis Georgi.In summary, Dayuanyin significantly attenuate LPS-induced ALI via reducing productions of pro-inflammatory mediators, decreasing the level of complement 3, 5a and increasing productions of anti-inflammatory mediators. DYY-4 and DYY-5 might play the major treatment role in ALI.
