Objective:The objective of this study was to investigate whether provide hydrogen sulfide could relief chemotherapy-induced neuropathic pain.Discuss the mechanism of how chemotherapy could induce neuropathic pain and the mechanism of what role hydrogen sulfide plays of relieving chemotherapy-induced neuropathic pain.Methods:Neuropathic pain was induced in mice by administration of vincristine for two cycles.At the same time administration of NaHS,Mechanical allodynia was evaluated by measuring the hind paw withdrawal responses to see whether neuropathic pain could be relieved.Meanwhile the expression of GFAP(astrocytic activation marker),Iba-1(microglial activation marker)and IL-1β(proinflammatory cytokines)were evaluated using qPCR and Western blot assay.Results:The hind paw withdrawal responses test showed that 50% paw withdrawal threshold of VCR group significantly decreased,the threshold of VCR+NaHS group was above VCR group.The expression level of GFAP and IL-1βsignificantly increased but after using NaHS,the level lowered in the result of qPCR and Wetern blot.The expression level of Iba-1 changed a little.Conclusions:Chemotherapy induced neuropathic pain was mainly via astrocytic activation in spinal cord.Hydrogen sulfide could relieve chemotherapy-induced neuropathic pain. Hydrogen sulfide could inhibit the activation of astrocytics,meanwhile could lower the expression of IL-1β in spinal cord.
